Water quality of aquatic habitats is an important determinant of female mosquito oviposition and successful larval development. This study examined the influence of environmental covariates on Anopheles arabiensis mosquito abundance in the Mwea Irrigation Scheme, Central Province of Kenya, prior to implementation of a malaria vector control program. Experimental rice plots were used to examine the environmental covariates responsible for regulating abundance and diversity of the aquatic stages of malaria vectors. Mosquito larval sampling and water quality analysis were done weekly from the flooding stage to the rice maturation stage. Sampling for mosquito larvae was conducted using standard dipping technique. During each larval collection, environmental covariates such as pH, temperature, conductivity, salinity, dissolved oxygen, water depth, and rice stage were measured. Anopheles arabiensis larval density was highest between 1 wk before transplanting and 4 wk after transplanting with peaks at weeks 0, 3, and 8. The fluctuation in values of the various environmental covariates showed characteristic patterns in different rice growth phases depending on the changes taking place due to the agronomic practices. Using a backward linear regression model, the factors that were found to be associated with abundance of An. arabiensis larvae at any of the rice growing phases included the following: dissolved oxygen, pH, turbidity, water depth, rice height, number of rice tillers, salinity, conductivity, and temperature. The environmental covariates associated with abundance of An. arabiensis were associated with early vegetative stage of the rice growth. For effective control of developmental stages of mosquito larvae, the application of larvicides should be done at the vegetative stage and the larvicides should persist until the beginning of the reproductive stage of the rice.

Rift Valley fever (RVF) continues to be a significant problem in Kenya as well as in Egypt, Yemen, and Saudi Arabia. In order to determine the ability of Kenyan mosquitoes to transmit RVF virus (RVFV), we collected mosquitoes in the Lake Naivasha region of Kenya and evaluated them for their potential to transmit RVFV under laboratory conditions. After feeding on a hamster (Mesocricetus auratus) with a viremia of 10^9.7 plaque-forming units of virus/ml of blood, Culex zombaensis were highly susceptible to infection with RVFV, with 89% becoming infected. In contrast, Cx. quinquefasciatus that were fed on the same hamsters were marginally susceptible, with only 20% becoming infected. Differences in percentages of mosquitoes that developed a disseminated infection were equally disparate, with 55% and 8%, for Cx. zombaensis and Cx. quinquefasciatus, respectively. Forty-eight percent of the Cx. zombaensis with a disseminated infection that fed on a susceptible hamster transmitted virus by bite, indicating a moderate salivary gland barrier. However, the presence of a salivary gland barrier could not be determined for Cx. quinquefasciatus because none of the 18 mosquitoes that took a 2nd blood meal had a disseminated infection. These studies illustrate the need to identify the ability of individual mosquito species to transmit RVFV so that correct decisions can be made concerning the application of appropriate control measures during an outbreak.

To determine their relative roles in transmission of dengue virus (DENV) in the Torres Strait region of northern Australia, we examined infection and dissemination of a sympatric strain of dengue virus type 2 (DENV-2) in Aedes scutellaris, Ae. albopictus, and Ae. aegypti. In experiments using membrane feeders for virus exposure, infection rates were 83% and 43% for Ae. scutellaris and Ae. aegypti, respectively. Salivary gland infection rates for both species were 43%. In experiments using pledgets for virus exposure, infection rates for Ae. aegypti, Ae. scutellaris, and Ae. albopictus were 68%, 55%, and 37%, respectively. Aedes albopictus exhibited the greatest barriers to infection with only 7% tested developing a salivary gland infection, compared to 42% and 24% of Ae. aegypti and Ae. scutellaris, respectively. These results suggest that Ae. scutellaris may have been responsible for DENV transmission on Torres Strait islands, where Ae. aegypti does not occur. In contrast, Ae. albopictus may not be an important vector of DENV-2 from the Torres Strait.

We conducted a laboratory evaluation of the ability of commercial antigen-capture assays, the Rapid Analyte Measurement Platform (RAMP®) and the VecTest® wicking assay, as well as Real Time reverse transcriptase–polymerase chain reaction (RT-PCR, Taqman) and Vero cell plaque assay to detect West Nile virus (WNV) in large mosquito pools. Real-Time PCR (Taqman) was the most sensitive, detecting WNV ribonucleic acid (RNA) in 100% of samples containing a single infected mosquito in pool sizes of up to 500 mosquitoes. Mosquito body tissues minimally impacted the ability of Real Time RT-PCR to detect WNV in a pool size of 500, reducing sensitivity by 0.6 log₁₀ plaque-forming units (PFU)/ml. Vero cell plaque assay detected live virus from a single infected mosquito in 100% of pools containing up to 200 mosquitoes, but was unreliable at larger pool sizes. VecTest detected 100% of positive pools containing 50 mosquitoes with 5.8 log₁₀ PFU/ml virus, 100 mosquitoes with 5.9 log₁₀ PFU/ml, and 200 mosquitoes with 5.2 log₁₀ PFU/ml. The RAMP assay detected 100% of positive pools containing 50 mosquitoes with 3.3 log₁₀ PFU/ml virus, 100 mosquitoes with 3.7 log₁₀ PFU/ml, and 200 mosquitoes with 4.0 log₁₀ PFU/ml. Results indicate that WNV can be reliably detected by all 4 assays in pools of mosquitoes exceeding 50 specimens, though there is some loss of sensitivity with very large pool sizes.

Geographic Information Systems (GIS) analysis of 34 forecasted high West Nile virus (WNV) risk communities in British Columbia (BC), Canada was useful to assess feasibility and planning of the operational logistics of an emergency spray event in advance of a WNV outbreak. The geographic coverage and operational time required to perform ground- and aerial-based ultra-low volume (ULV) adulticiding were calculated using GIS. The mean geographic coverages of the ground-, aerial-, and combination of ground- and aerial-based adulticiding strategies were 39%, 61%, and 69%, respectively. The driving distance, driving time, and number of treatment nights required to perform ground-based spraying of an entire community were also calculated. Due to the large variability of treatment coverage estimates within and among the communities, no single treatment method was identified as the best strategy for province-wide ULV adulticiding in BC. Instead, the strategy for each community should be examined individually with local knowledge and expertise.

Eliminating infected female mosquitoes by aerial applications of ultra-low volume adulticides is the intervention strategy currently recommended to interrupt the epidemic transmission of encephalitis viruses, including West Nile. The current research optimized pyrethrin formulations and evaluated their efficacy in the desert environment of the Coachella Valley, Riverside County, California. After seven trials during 2004, a 1:2 by volume mixture of Pyrenone 25-5 in BVA oil optimized particle size, droplet descent to ground level, and kill of sentinel mosquitoes. Three subsequent experiments used 3 aerial applications of the 1:2 Pyrenone 25-5:BVA oil mixture on alternate nights to suppress Culex tarsalis Coquillett host-seeking abundance over a 1-square-mile target area. Mortality patterns among caged sentinel mosquitoes varied among sites and replicate sprays, indicating variable particle dispersion at ground level within the target area. In addition, mortality was observed for sentinels up to 1 mile downwind from the target area, indicating considerable particle drift. Geometric mean abundance of host-seeking Cx. tarsalis females collected at dry ice–baited traps within each of 3 sprayed and 2 unsprayed negative control strata varied similarly over time, indicating that our sprays minimally impacted the target population or that drift combined with other factors led to widespread area control. Experiments during March and June when recruitment rates were minimal showed general area-wide suppression of abundance following spray, whereas an experiment during September when recruitment rates were high from newly flooded marshes failed to prevent an area-wide increase in abundance. Clearly additional research is needed to standardize the efficacy of aerial applications of pyrethrins in hot dry desert environments.

Eight Latin American strains of Aedes aegypti were evaluated for resistance to 6 organophosphates (temephos, malathion, fenthion, pirimiphos-methyl, fenitrothion, and chlorpirifos) and 4 pyrethroids (deltamethrin, lambdacyhalothrin, betacypermethrin, and cyfluthrin) under laboratory conditions. In larval bioassays, temephos resistance was high (resistance ratio [RR₅₀], ≥10×) in the majority of the strains, except for the Nicaragua and Venezuela strains, which showed moderate resistance (RR₅₀, between 5 and 10×). The majority of the strains were susceptible to malathion, fenthion, and fenitrothion. However, resistance to pirimiphos-methyl ranged from moderate to high in most of the strains. Larvae from Havana City were resistant to 3 of the pyrethroids tested and moderately resistant to cyfluthrin. The Santiago de Cuba strain showed high resistance to deltamethrin and moderate resistance to the other pyrethroids (lambdacyhalothrin, betacypermethrin, and cyfluthrin). The rest of the strains were susceptible to pyrethroids, except for the Jamaica and Costa Rica strains, which showed moderate resistance to cyfluthrin, and Peru and Venezuela, which showed resistance to deltamethrin. Adult bioassays showed that all the strains were resistant to dichlorodiphenyl-trichloroethane and to the majority of pyrethroids evaluated. The use of the synergists S,S,S,-tributyl phosphorotrithioate and piperonil butoxide showed that esterase and monooxygenases played an important role in the temephos, pirimiphos-methyl, and chlorpirifos resistance in some strains. Biochemical tests showed high frequencies of esterase and glutathione-S-transferase activity; however, the frequency of altered acetylcholinesterase mechanism was low. The polyacrylamide electrophoresis gel detected the presence of a strong band called Est-A4. Insecticide resistance in Ae. aegypti is a serious problem facing control operations, and integrated control strategies are recommended to help prevent or delay the temephos resistance in larvae and pyrethroids resistance in adults.

Two 6-wk trials were conducted in 28-m² earthen ponds to compare the efficacy of the arroyo chub, Gila orcutti, to the mosquitofish, Gambusia affinis, as a biological control agent for mosquitoes and a possible replacement for the mosquitofish in sensitive watersheds of southern California. The mosquitofish population growth rate was 1.73 times greater than the arroyo chub population growth rate; however, greater reproduction by the mosquitofish did not result in significantly better reduction of mosquitoes than was provided by the comparatively small populations of arroyo chub. On average across a 6-wk study in the spring, both larvivorous fishes reduced the abundance of 3rd and 4th instars by 4- to 5-fold compared to that observed in the control ponds that lacked fish but contained few invertebrate predators. The abundance of nontarget microinvertebrates in ponds containing the mosquitofish was only 7% of that in ponds containing the arroyo chub during the summer, but did not differ significantly between the fish species treatments when zooplankton was comparatively more abundant during the spring. Even though the number of individuals produced by each fish species during 6 wk in the spring was greater than for fish stocked in the summer, species-specific population growth rates in the spring study (individuals/individual/d; mosquitofish, 0.077; arroyo chub, 0.044) were only slightly higher than in the summer (individuals/individual/d; mosquitofish, 0.068; arroyo chub, 0.039) indicating that differences in the number of fish stocked contributed primarily to the differences in final population size between spring and summer studies. The arroyo chub is native to the South Coastal drainages in California and should be considered as a viable alternative to the mosquitofish for integrated mosquito management programs in riverine wetlands and sensitive watersheds of southern California.

The emergence of West Nile virus in the United States renewed vigilance for mosquito-borne diseases and rejuvenated mosquito surveillance activities in Washington State. As part of these activities, the Zoonotic Disease Program, Washington Department of Health, and the United States Army Center for Health Promotion and Preventive Medicine-West collaborated to produce this revision of the distribution of mosquitoes in Washington State. Data from these organizations, the US Air Force, and county surveillance records, as well as published literature and mosquito collections, were used to develop this statewide mosquito distribution checklist.

The 17th Annual Latin American symposium presented by the American Mosquito Control Association (AMCA) was held as part of the 73rd Annual Meeting in Orlando, FL, in April 2007. The principal objective, as for the previous 16 symposia, was to promote participation in the AMCA by vector control specialists, public health workers, and academicians from Latin America. This publication includes summaries of 57 presentations that were given orally in Spanish or presented as posters by participants from 6 countries in Latin America, Puerto Rico, and the USA. Topics addressed in the symposium included results from chemical and biological control programs and studies; studies of insecticide resistance; and population genetics, molecular, ecological, and behavioral studies of vectors of dengue (Aedes aegypti and Ae. albopictus), Venezuelan equine encephalitis (Culex spp.), West Nile viruses (Cx. nigripalpus and Cx. quinquefasciatus), malaria (Anopheles spp.), leishmaniasis (Lutzomyia), and onchocerciasis (Simulium).

Aedes albopictus were collected in the French military camp of Libreville, Estuaire Province, Gabon, from January to March 2007 by human landing catches during an entomological evaluation of malaria transmission. Inspection of potential larval habitats within and outside the camp showed that Ae. albopictus was found only in artificial containers (discarded tires and small water containers). Associated species of mosquito larvae were Ae. aegypti (L.) and Culex quinquefasciatus. At the same time, Ae. albopictus adults and larvae were also collected from discarded tires in Tcheungue near Port Gentil, Ogoue Maritime Province. Ae. albopictus seems to be established in this part of Gabon's littoral. Further studies are necessary to investigate the extension of Ae. albopictus establishment throughout the country.

Females of Culex coronator were collected in Mobile, AL, during surveillance to direct mosquito control operations following Hurricane Katrina. A total of 249 specimens of this species were collected in light traps at 15 widely separated sites in the county. These represent the first confirmed specimens of Cx. coronator in Alabama.

The first confirmed record of Psorophora ciliata from Maine was made at South Berwick, ME, from a carbon dioxide–baited light trap collection on July 25, 2006. Included are collection site data and species bionomics.

A Bacillus sphaericus highly toxic to 4th-stage Culex quinquefasciatus was isolated from sediment samples collected at Winter Beach marsh in Indian River County, Florida. This isolate, named WBM 1-1-13, showed significantly higher toxicity compared with strain 2297. WBM 1-1-13 is equivalent, though better initially, to the toxicity of strain 2362, the active ingredient of VectoLex®. Furthermore, the Winter Beach marsh isolate produced more Bin per unit medium than strain 2362, which suggests that this new isolate could be useful for Culex control.

The efficacy of the larvivorous fish Poecilia reticulata against mosquito larvae was monitored in a drain at Bharat Heavy Electricals Limited, Hardwar, India. The water was polluted and the water flow was in some way impeded. Poecilia reticulata failed to feed on Culex quinquefasciatus larvae in this drain. Laboratory experiments also confirmed the inefficacy of P. reticulata as a predator of Cx. quinquefasciatus larvae during the first 24 h. Significant differences in the efficacy of P. reticulata against Cx. quinquefasciatus were recorded between polluted water and drinking water. Poecilia reticulata preferred to feed on other available food present in the polluted water rather than on Cx. quinquefasciatus larvae. This was verified by the identification of plankton in the gut content of the fish and by the high density of plankton present in the polluted water.

A new cage was designed to study caged mosquitoes in ultra-low volume insecticide spray studies in the field. Insecticide droplets may enter the cage from any direction because all surfaces are mesh. Cages can be collapsed in seconds, and reassembly is quick and simple. A 26-cm-diam cage can be constructed for less than US$6.00.

Culex pipiens, Cx. restuans, and Cx. salinarius play important and most likely different roles in transmission of West Nile virus (WNV) in the northeastern United States. While Cx. pipiens and Cx. restuans are considered the main enzootic vectors of WNV, Cx. salinarius may be involved in epizootic cycles due to its broader host preferences. Accurate morphological identification of field-collected Culex specimens may be difficult, and therefore the New York State Department of Health arbovirus surveillance program allows combined Cx. pipiens and Cx. restuans pools to be tested for WNV. We have developed a modified and improved DNA isolation protocol using proteinase K digestion without traditional mosquito trituration and nucleic acid extraction to permit high-throughput screening of a large number of Culex specimens for species identification using polymerase chain reaction (PCR). This method utilizes a 96-well–plate format and a novel 1-step crude extraction procedure using proteinase K to obtain genomic DNA template from 1 mosquito leg in sufficient quantity for at least 2 standard 50-µl PCR reactions. Proteinase K digestion of legs from individual Culex mosquitoes was performed and used for PCR amplification with previously described species-specific ribosomal DNA primers. Using these rDNA primers, our modified proteinase K method successfully identified 91% to 100% of the Culex samples.

An insect aspirator was modified to remove mosquitoes that entered an animal-baited experimental cage within a cage. The modified aspirator is easy to maneuver inside tight spaces, powerful enough to aspirate mosquitoes but not remove scales or fluorescent marking powders, and will run continuously for at least 45 min.
