Tests were conducted to determine the potential for high hydrostatic pressure (HPP) to control codling moth, Cydia pomonella (L.), and western cherry fruit fly, Rhagoletis indifferens Curran. Apples (Malus spp.) with codling moth larvae or eggs were treated at 24 and 72 h, respectively, after infestation at a series of pressures between 14,000 and 26,000 pounds per inch² (psi). Survivorship was determined the next day for larvae and after 10 d for eggs. Codling moth eggs were more tolerant of HPP treatment than larvae. Mortality of larvae was 97% at 22,000 psi, whereas mortality of eggs at this dose was 29% and not significantly different from the untreated controls. In a second study, no codling moth eggs hatched at any high pressure treatment between 30,000 and 80,000 psi, indicating these pressures were lethal. Various stages of western cherry fruit fly were treated at pressures from 10,000 to 45,000 psi, and survivorship was determined after 24 h. Eggs and third instars were more tolerant of HPP than the first and second instars. Mortality was 100% in western cherry fruit fly eggs and larvae at pressures ≥25,000 psi. Apple and sweet cherry quality after high pressure treatment was poor, but high pressure may have applications to control quarantine pests in other fruits.

In Australia, the brackish water mosquito Verrallina funerea (Theobald) (Diptera: Culicidae) is a serious pest and vector of Ross River and Barmah Forest viruses in coastal areas in the northeast of the continent. We report on the first extensive evaluation of the efficacy of various pesticides against this species, including laboratory dose–response assays and small plot field trials. In the laboratory, Ve. funerea was susceptible to temephos (Abate 100E) (lethal concentration, 95% endpoint [LC₉₅] of 1.651 ppb), Bacillus thuringiensis subsp. israelensis de Barjac (Bti) (VectoBac 12AS) (LC₉₅ of 0.033 ppm), s-methoprene (ProLink Liquid Larvicide [PLL]) (emergence inhibition, 95% endpoint [EI₉₅] of 2.647 ppb), and technical grade pyriproxyfen (EI₉₅ of 0.227 ppb). In contrast, Ve. funerea was highly refractory to Bacillus sphaericus Neide (VectoLex WG) (LC₉₅ of >855.3 ppm). The survival of adults exposed as larvae to sublethal doses of s-methoprene was reduced for those doses greater than the EI₅₀, although sex ratios were unaffected. Field trials demonstrated that Ve. funerea was susceptible to Bti (VectoBac G at 7 kg/ha) and s-methoprene (ProLink ProSand at 4 kg/ha), with these products giving >99 and >98% control, respectively. We also observed a significant increase in immature mortality in s-methoprene-treated pools, suggesting that s-methoprene field trials based solely on emergence inhibition of pupae may underestimate the efficacy of this insecticide.

A discriminating irradiation dose of 150 gray (Gy) was used to determine the most tolerant immature stages of Opogona sacchari (Bojer) (Lepidoptera: Tineidae). Based on adult emergence, early and late pupae were determined to be the most tolerant stages, and they were significantly more tolerant than eggs, neonate larvae, and larvae that were 1, 2, or 3 wk old. Irradiation treatment of eggs, neonates, 1-wk-old larvae, 2-wk-old larvae, 3-wk-old larvae, early pupae, and late pupae at 150 Gy resulted in a 96, 96, 95, 73, 61, 8, and 9% reduction in adult emergence, respectively. Pupae were treated with irradiation doses between 60 and 400 Gy. Emergence to the adult stage was significantly reduced by irradiation, averaging 90% in experimental controls and 29% in the 400-Gy treatment. Egg production was also reduced by irradiation, although the average age of pupae at the time of irradiation had a larger effect on fecundity. In total, 2,527 pupae treated with 120 Gy eclosed and produced 47,221 eggs and three F₁ larvae. In the 150-Gy treatment, 2,927 adults in total emerged from the 4,626 insects treated as pupae. These adults laid 62,878 eggs, none of which hatched. We conclude that a minimum dose of 150 Gy should be sufficient for sterilization of immature O. sacchari infesting export commodities.

Genetic diversity within and among three wild-type natural populations and one melanic laboratory population of Mythimna separata (Walker) (Lepidoptera: Noctuidae) were evaluated using amplified fragment length polymorphism (AFLP) analysis. Although extensive genetic diversity occurs among individuals from different geographic populations (P = 54.5%, h = 0.209, I = 0.305), the majority of the genetic diversity is within populations and not between populations (G_ST = 0.172), indicating high gene flow (N_M = 2.403) and suggesting that M. separata in northern China are a part of a single large metapopulation. Genetic diversity in the natural populations was significantly higher than that in the melanic laboratory population (with P = 43.4% versus P = 25.9%, h = 0.173 versus h = 0.086, and I = 0.251 versus I = 0.127), suggesting that the melanic laboratory population is narrowly genetic-based and genetically uniform. Genetic similarities based on AFLP data were calculated, and cluster analysis was preformed to graphically display groupings between individuals and populations. Individuals from the same region were not grouped together in cluster analysis of three natural populations, whereas melanic individuals from laboratory population were grouped together very well. Four subpopulations were clustered into two broad groups. Melanic laboratory population became a single group, which had apparent differentiation from the other group in which three natural subpopulations were included. These results indicated that although high genetic variability existed among the individuals of natural populations, there was little genetic differentiation among three geographic populations that could be explained by the effects of the long distance migration of the oriental armyworm in China enhanced the level of gene flow. Influences of migration on the genetic polymorphism and differentiations that make a significant contribution to evolution in this insect are reviewed.

Penicillin and other antibiotics are routinely incorporated in insect culture media. Although culturing insects in the presence of antibiotics is a decades-old practice, antibiotics can exert deleterious influences on insects. In this article, we test the hypothesis that one of the effects of dietary penicillin is to increase oxidative stress on insects. The effects of penicillin on midgut concentrations of the oxidative stress indicator malondialdehyde (MDA) and on midgut antioxidant enzyme (superoxide dismutase [SOD], catalase [CAT], glutathione S-transferase [GST], and glutathione peroxidase [GPx]) and transaminases (alanine aminotransferase and aspartate aminotransferase) activities in greater wax moth, Galleria mellonella (L.), were investigated. The insects were reared from first instars on artificial diets containing 0.001, 0.01, 0.1, or 1.0 g penicillin per 100 g of diets. MDA content was significantly increased in the midgut tissues of each larval instar reared in the presence of high penicillin concentrations. Activities of antioxidant and transaminase enzymes did not show a consistent pattern with respect to penicillin concentrations in diet or age of larvae. Despite the increased penicillin-induced oxidative stress in gut tissue, antioxidant and transaminase enzymes did not correlate with oxidative stress level or between each other in larvae of other age stages except for the seventh instar. We found a significant negative correlation of MDA content with SOD and GST activities in seventh instars. SOD activity was also negatively correlated with CAT activity in seventh instars. These results suggest that exposure to dietary penicillin resulted in impaired enzymatic antioxidant defense capacity and metabolic functions in wax moth larval midgut tissues and that the resulting oxidative stress impacts midgut digestive physiology.

Diabrotica barberi Smith & Lawrence and Diabrotica virgifera virgifera LeConte (Coleoptera: Chrysomelidae) are serious pests of maize, Zea mays L. To reduce the amount of toxicants released into the environment, the Agricultural Research Service implemented a 5-yr (1997–2001) areawide pest management program in five geographic locations, including one in South Dakota. The objective was to use integrated pest management tactics to suppress adult Diabrotica populations over a broad geographic area by using aerially applied semiochemical-baited insecticides. Suppressed populations theoretically should reduce oviposition, limit larval feeding damage to maize roots, and result in fewer beetles emerging in subsequent years. We used emergence cages, sticky traps, and CRW lure traps to monitor adult D. barberi and D. v. virgifera populations. We sampled for Diabrotica eggs, and we determined damage to maize roots. We sampled in several maize fields (control) located near the areawide site. The baited insecticides were effective in reducing adult populations 1 and 2 wk after application, and most remained low for the duration of the maize growing season. Fewer beetles were captured in both sticky and lure traps in the areawide site than in the control site. With a few exceptions, egg counts, adult emergence, and maize root damage were similar between the areawide and control sites; however, maize roots had greater fresh weight in the control site. Although not all goals were accomplished, when considering the amount of toxicant released into the environment, using semiochemical-baited insecticides to suppress adult pest Diabrotica populations seems to be an effective areawide management tool.

The guild of stem-infesting insect pests of cultivated sunflower, Helianthus annuus L., within the central Plains is a concern to producers, chiefly due to losses caused by plant lodging from the sunflower stem weevil, Cylindrocopturus adspersus (LeConte) (Coleoptera: Curculionidae), and Dectes texanus texanus LeConte (Coleoptera: Cerambycidae). The incidence of a root boring moth, Pelochrista womonana (Kearfott) (Lepidoptera: Tortricidae), also has increased. Experiments were conducted in Kansas during 2000–2001 to investigate the effect of irrigation timing and intensity on densities of C. adspersus, D. texanus, and P. womonana larvae within cultivated sunflower stalks. Supplemental soil moisture provided by irrigation during the growing season increased both seed yield and oil content, and it reduced insect densities of the sunflower stem weevil and P. womonana in the sunflower stalk. Results showed that ensuring adequate moisture during the growing season can assist in reducing stem-infesting insect densities, revealing an additional advantage of crop irrigation beyond improved sunflower productivity.

The influence of southern green stink bug, Nezara viridula (L.), adults (males and females) and fourth to fifth instars on cotton, Gossypium hirsutum L., boll abscission, seedcotton yield, fiber quality, and seed viability was evaluated in field studies conducted during 2004 and 2005. Cotton bolls representing several age classes ranging from 0–600 heat units were individually infested with a specific gender or life stage of southern green stink bug. Adults and nymphs induced abscission of bolls that accumulated 0–280 heat units after anthesis. Seedcotton yield was significantly lower in bolls infested with adults (males and females) and late instars through ≈500 heat units after anthesis. Southern green stink bug feeding on bolls significantly affected the physical fiber properties of micronaire (measure of fiber fineness or maturity), strength, uniformity, and fiber length. Discolored cotton lint in the stink bug-infested bolls was more common than in noninfested bolls. Seed germination and development of normal seedlings for seed harvested from stink bug-infested bolls that accumulated ≤500 heat units beyond anthesis were significantly lower compared with noninfested bolls. No significant differences in boll abscission, yield, fiber quality, and seed germination were detected between southern green stink bug males and females or between adults and fourth to fifth instars.

Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), or fall armyworm, is an important agricultural pest of several crops in the Western Hemisphere, including cotton (Gossypium L.). Two morphologically identical host strains of fall armyworm exist that differ in plant host use and habitat distribution. The corn-strain is a primary pest of corn, Zea mays L., whereas the rice-strain is the majority population infesting rice (Oryza spp.) and turfgrass (Cynodon spp.). With the increased use of Bacillus thuringiensis (Bt) toxin-expressing cotton varieties and the necessity of ensuring adequate refuge areas to prevent the spread of Bt toxin resistance, it is crucial to identify the alternative plant hosts available for the fall armyworm population infesting cotton. Stable isotope analysis combined with the molecular analysis of strain-specific markers was used to investigate whether one or both strains routinely develop on cotton grown in the Mississippi delta. We found that the majority of fall armyworm adults present during the early cotton growing season arose from C₄ plants (e.g., corn and sorghum, Sorghum vulgare Pers.) and that the only strain likely to be developing on cotton (a C₃ plant) in substantial numbers was the corn-strain. The population distribution patterns observed were consistent with corn providing an important refuge for the fall armyworm strain infesting cotton and suggested that late season populations in the Mississippi delta may be migrants from more northern corn areas.

Emerald ash borer, Agrilus planipennis Fairmaire (Coleoptera: Buprestidae), a phloem-feeding pest native to Asia, was identified in June 2002 as the cause of widespread ash (Fraxinus spp.), mortality in southeastern Michigan and Windsor, Ontario, Canada. Localized populations of A. planipennis have since been found across lower Michigan and in areas of Ohio, Indiana, Illinois, Maryland, and Ontario. Officials working to contain A. planipennis and managers of forestlands near A. planipennis infestations must be able to compare alternative strategies to allocate limited funds efficiently and effectively. Empirical data from a total of 148 green ash, Fraxinus pennsylvanica Marsh., and white ash, Fraxinus americana L., trees were used to develop models to estimate surface area of the trunk and branches by using tree diameter at breast height (dbh). Data collected from 71 additional F. pennsylvanica and F. americana trees killed by A. planipennis showed that on average, 88.9 ± 4.6 beetles developed and emerged per m² of surface area. Models were applied to ash inventory data collected at two outlier sites to estimate potential production of A. planipennis beetles at each site. Large trees of merchantable size (dbh ≥ 26 cm) accounted for roughly 6% of all ash trees at the two sites, but they could have contributed 55–65% of the total A. planipennis production at both sites. In contrast, 75–80% of the ash trees at the outlier sites were ≤13 cm dbh, but these small trees could have contributed only ≤12% of the potential A. planipennis production at both sites. Our results, in combination with inventory data, can be used by regulatory officials and resource managers to estimate potential A. planipennis production and to compare options for reducing A. planipennis density and slowing the rate of spread for any area of interest.

Insecticide bioassays were used to investigate resistance of Cydia pomonella (L.) (Lepidoptera: Tortricidae) to insecticides with various types of active ingredients. The efficacy baselines of selected insect growth regulators (fenoxycarb), insect growth inhibitors (diflubenzuron and teflubenzuron), organophoshorous insecticides (phosalone), and neonicotinoids (thiacloprid) against the eggs and first and fifth instars of sensitive laboratory strains of codling moth were determined. According to concentration–mortality baseline, 50% lethality concentration values and 90% lethality concentration values were determined for all the tested insecticides. The lethal concentration ratio quantified the relation between the efficacy of selected insecticides against fifth instars found by topical application and against first instars found by diet-treated bioassay. No difference was detected when the efficacy of technical grade diflubenzuron diluted in tetrahydrofuran and diflubenzuron in the formulated product Dimilin 48 SC diluted in water was compared. However, just before the application of insecticide, the integument of larvae must be treated with acetone. Two bioassays were used to monitor the resistance of codling moths collected in 2003–2005 in two apple (Malus spp.) orchards with different intensities of chemical control. Resistance ratios (RRs) to the tested insecticides were determined for both field populations of codling moth. For the population of codling moth from a commercial apple orchard in Velké Bílovice, cross-resistance to fenoxycarb, teflubenzuron, and phosalone was detected after the topical application of insecticides to fifth instars. The population of codling moth from Prague-Ruzyně was slightly resistant to phosalone and teflubenzuron. No resistance to diflubenzuron was detected in either tested population.

Field-based bioassays and residue profile analysis were used to determine the relative importance of lethal and sublethal effects of imidacloprid on adult Japanese beetle, Popillia japonica Newman, in blueberries, Vaccinium corymbosum L. Field-based bioassays assessed adult mortality and knockdown, and fruit and leaf injury from Japanese beetles exposed to 4-h and 7-d field-aged residues of imidacloprid, and the conventional insecticides azinphosmethyl and esfenvalerate. Azinphosmethyl and imidacloprid caused high levels of mortality when beetles were exposed to blueberry shoots with ripe fruit 4 h postapplication, and all compounds protected blueberry fruit and foliage from beetle feeding. Azinphosmethyl and esfenvalerate caused significant Japanese beetle mortality when adults were exposed to blueberry shoots 7 d postapplication, whereas imidacloprid residues caused effects that protected leaves, although not of ripe fruit. When beetles were exposed to shoots with immature green fruit, relatively more leaf feeding and mortality were observed, suggesting that earlier treatment timings may be most effective for systemic neonicotinoids. Japanese beetle mortality was highly correlated with imidacloprid fruit and leaf surface residues, whereas sublethal feeding deterrent effects were observed after the surface residues diminished. The value of the plant-insect-chemistry model for describing the spatial and temporal dimensions of insecticide modes of activity is discussed in terms of optimizing crop protection.

Bacillus thuringiensis serovar japonensis strain Buibui experimental formulations containing Cry8Ca1 δ-endotoxin were evaluated in the field for the control of oriental beetle, Anomala orientalis Waterhouse; Japanese beetle, Popillia japonica Newman; Asiatic garden beetle, Maladera castanea (Arrow); and European chafer, Rhizotrogus majalis (Razoumowsky) larvae infesting turfgrasses. In 17 fall applications, control of A. orientalis and P. japonica was achieved at rates as low as 100 g of toxin per ha; however, rates of 301 or 593 g/ha were consistently more effective. A. orientalis larvae were the most susceptible followed by P. japonica, R. majalis, and M. castanea. Two spring applications of 120 g of toxin per ha were not effective against oriental or Japanese beetles.

Standard soil application methods (granular and drench) were compared with novel methods (tablet, stick soak, and root dip) for efficacy and duration in hybrid poplar (Populus spp.) against adult and larval cottonwood leaf beetle, Chrysomela scripta F. (Coleoptera: Chrysomelidae). Beetle feeding can kill saplings and significantly damage trees by reducing tree height, diameter, and biomass. Tablets offer lower risk to applicators and beneficial insects, because insecticides do not need to be measured and sprayed. In field- and container-grown plants, standard granular and drench application methods had similar efficacy and duration compared with tablets. In field-grown plants, adult and larval survivorship was reduced for 12 mo with the two highest rates of tablet (0.25× and 0.5×) treatments, and in container-grown plants, with all rates of tablet (0.25×, 0.5×, and 1×) treatments that were used. Two other novel application methods, stick soak and root dip, offer new methods for protecting vulnerable transplants in nursery propagation. In container-grown plants, adult survivorship was reduced for 8 mo and larval survivorship for 12 mo for all rates of stick soak (0.5×, 1×, and 2×) and all rates of root dip (1×, 2×, and 4×) treatments. Literature searches revealed little data on the efficacy and duration of soil-applied imidacloprid for trees, even though it is the primary insecticide used for defoliators and some borers in landscape and in nurseries for field and container production.

The dispersal behavior of flower thrips was studied during two field seasons within blueberry (Vaccinium spp.) plantings in Florida and southern Georgia. A “shake and rinse” technique used to extract thrips from inside the blueberry flowers was not significantly different from the conventional dissecting technique, but the time taken to complete the extraction of thrips was significantly shorter. Overall, the highest concentration of thrips was captured inside the canopy of blueberry bushes. Using a grid of traps to monitor the dispersal of thrips during the blueberry flowering season, we analyzed their dispersion with graphical and analytical methods to determine and describe their distribution within blueberry plantings. Thrips began to form “hot-spots” 5–7 d after bloom initiation. A hot-spot is defined as a large number of thrips concentrated in a small area of the field, whereas the rest of the field has a low population. The behavior of the population inside these hot-spots fit a Gaussian tendency and a regression was conducted to describe this tendency. Green’s and Standardized Morisita’s indices were used to determine thrips level of aggregation. Results showed significantly aggregated populations of thrips in both years. Formation of hot-spots in blueberry plantings seemed to be random. However, the formation of hot-spots was higher in places where more than seven thrips per day were captured on sticky traps, 5 to 7 d after the bloom begins. With these results, producers will be able to monitor thrips populations and locate and manage hot-spots before they become a more serious a problem on blueberry farms.

Although the introduced lady beetle Harmonia axyridis (Pallas) (Coleoptera: Coccinellidae) is an important predator of aphids in a variety of crop systems during the growing season, it is often a pest in fall and winter when it enters buildings seeking overwintering sites. One of the primary recommendations for managing this annual influx is to prevent beetle entry by caulking or otherwise filling potential entry points in buildings. The goal of this study was to determine how small a gap the beetles are able to enter in choice and no-choice studies by experimentally exploiting their behavioral tendency to seek dark shelters at cool temperatures. Within the size range of adults collected in central North Carolina in 2003 (1.99–3.29 mm body height), no beetles entered a 2-mm access during no-choice experiments. Most (83%) entered a 3-mm gap; those failing to cross the 3-mm threshold were significantly larger than those that traversed it. In choice experiments, 98.2% of beetles entered shelters. As in the previous study, no beetles entered shelters with 2-mm gaps. Significantly fewer were found in shelters with 3-mm entrances than in those with 4- or 5-mm access; beetles that entered 3-mm gaps were significantly smaller than the remainder of the test population. Although no H. axyridis crossed a 2-mm threshold in either experiment, a gap of this size may nonetheless allow admission if it has flexible borders (e.g., foam weather stripping); beetles were observed attempting forced entry into too-small crevices.

In northern Italy (Emilia-Romagna region), integrated pest management has been used for several years against pear psylla, Cacopsylla pyri L. (Hemiptera: Psyllidae), a relevant pest of pear (Pyrus spp.) trees. After the outlawing of amitraz in 2005, the most common active ingredient involved is abamectin, a mixture of avermectin B_1a and avermectin B_1b. After the development of C. pyri resistance to azinphos methyl in southern France, we evaluated, by topical application, the different sensitivities to abamectin on C. pyri populations collected in orchards from Emilia-Romagna, where different field strategies were used, with alternative success in terms of pest management. The LC₅₀ values ranged between 1.61 and 28.37 mg/liter, and they revealed variations more related to collection time than to field strategies. The failure of abamectin treatments against C. pyri in some Emilia-Romagna locations is probably unrelated to resistance development, but rather it is related to incorrect pest defense management, which could interfere with pest parasitoids and predators.

Transgenic crops producing insecticidal toxins are now widely used to control insect pests. The benefits of this method would be lost if resistance to the toxins spread to a significant proportion of the pest population. The primary resistance management method, mandatory in the United States, is the high-dose/refuge strategy, requiring toxin-free crops as refuges near the insecticidal crops, and the use of toxin doses sufficiently high to kill insects heterozygous for a resistance allele, thereby rendering resistance functionally recessive. We propose that mass-release of harmless susceptible (toxin-sensitive) insects could substantially delay or even reverse the spread of resistance. Mass-release of such insects is an integral part of release of insects carrying a dominant lethal (RIDL), a method of pest control related to the sterile insect technique. We show by mathematical modeling that specific RIDL strategies could form an effective component of a resistance management strategy for plant-incorporated protectants and other toxins.

We determined effects of aerial sprays of the insect growth regulator pyriproxyfen on sweetpotato whitefly, Bemisia tabaci (Gennadius) (B biotype), in Arizona cotton (Gossypium spp.) fields. We measured survival for males and females from a susceptible strain and a laboratory-selected resistant strain, as well as for hybrid female progeny from crosses between the strains. Insects were exposed directly to pyriproxyfen sprays in the field or indirectly in the laboratory by rearing them on sprayed leaves collected from the field. In all tests, survival was higher for the resistant strain than the susceptible strain, but did not differ between sexes in each strain. Survival to the adult stage did not differ between eggs and nymphs directly exposed to sprays. For susceptible and hybrid individuals, survival was lower on leaves collected the day of spraying than on leaves collected 2 wk after spraying. In contrast, survival of resistant individuals did not differ based on the timing of exposure. Dominance of resistance to pyriproxyfen depended on the type of exposure. Resistance was partially or completely dominant in direct exposure bioassays and on leaves collected 2 wk after spraying (h > 0.6). Resistance was partially recessive on leaves collected the day of spraying (mean h = 0.34). Rapid evolution of resistance to pyriproxyfen could occur if individuals in field populations with traits similar to those of the laboratory-selected strain examined here were treated intensively with this insecticide.

The soybean aphid, Aphis glycines Matsumura (Hemiptera: Aphididae), is a worldwide pest of soybean, Glycine max (L.) Merr. Studies to find control methods were initiated in 2000 when it was first detected in North America. A. glycines can reduce yields by as much as 50%, and it is the vector of several viral diseases. A. glycines removes phloem sap, which can result in a reduction of chlorophyll content. Quantification of chlorophyll loss caused by A. glycines feeding on soybean is of vital importance. The SPAD-502 chlorophyll meter is a device that has been used to measure chlorophyll loss caused by nonchewing insects. Chlorophyll loss was studied in no-choice tests on the infested and uninfested leaves of a susceptible check (KS4202). The minimum combined number of days and aphids needed to detect significant chlorophyll loss was 30 aphids confined for 10 d. In a similar experiment, seven resistant entries and two susceptible checks were evaluated. There was no significant chlorophyll reduction between infested and uninfested leaves of five of the resistant entries (K1621, K1639, Pioneer 95B97, Dowling, and Jackson). Percentage of loss of chlorophyll in the susceptible checks was ≈40%; Jackson and Dowling had a significantly lower percentage loss (13 and 16%, respectively) compared with the susceptible checks. The percentages of chlorophyll loss of K1621, K1639, and Pioneer 95B97 were not statistically different from the percentage of loss of Jackson.

RESUMEN El áfido de la soya, Aphis glycines Matsumura (Hemiptera: Aphididae), es una plaga de la soya, Glycine max (L.) Merr. En el año 2000 SE encontró A. glycines en Norte América, y desde entonces, SE iniciaron estudios para encontrar métodos de control. Aphis glycines reduce el rendimiento en un 50%, y transmite virus. Aphis glycines extrae el floema de las hojas lo cual puede tener efectos en la fisiología de la soya como la reducción de clorofila. Por lo tanto, la evaluación de las pérdidas de clorofila en soya causadas por A. glycines es de mucha importancia. El medidor SPAD-502 SE usa para cuantificar pérdidas de clorofila causadas por insectos no masticadores. Se estudiaron las pérdidas en hojas infestadas y no-infestadas del testigo susceptible KS4202. Se descubrió que el número mínimo de áfidos necesario para observar pérdidas significativas de clorofila es de 30 áfidos confinados por 10 días. En un experimento análogo, siete genotipos resistentes y dos susceptibles fueron evaluados. De los genotipos resistentes, cinco (K1621, K1639, Pioneer 95B97, Dowling y Jackson) no mostraron reducción en su contenido de clorofila en hojas infestadas y no-infestadas. El porcentaje de pérdida de los genotipos susceptibles fue del 40% aproximadamente; Jackson y Dowling tuvieron porcentajes de perdida (13 y 16%, respectivamente) significativamente más bajos que los testigos susceptibles. Sin embargo, los porcentajes de K1621, K1639 y Pioneer 95B97 no fueron estadísticamente diferentes al porcentaje de pérdida de Jackson.

Plum curculio, Conotrachelus nenuphar (Herbst) (Coleoptera: Curculionidae), is an important apple, Malus domestica Borkh., pest that significantly hinders sustainable apple production in eastern North America. The potential for host plant resistance to plum curculio among apple germplasm has never been rigorously evaluated. Thus, studies were conducted to assess the susceptibility of a number of exotic and domestic Malus accessions housed at the USDA Plant Genetic Resources Unit (PGRU) “core” collection in Geneva, NY. Contrary to earlier published reports and promising data from a field assessment in 2005, these results suggest that there is probably little potential for genetic resistance to plum curculio among the Malus germplasm collection evaluated. More specifically, four Malus hybrid selections that have previously been released with claims of plum curculio resistance were shown to be susceptible to plum curculio attack. Because there are additional accessions housed at PGRU outside of the core collection that are currently classified as resistant, further studies are necessary to evaluate the true resistance qualities of these releases. It is also important to clarify such discrepancies in both the USDA online Germplasm Resources Information Network and in the horticultural literature. Although other Malus species exhibited some variability in fruit susceptibility, none could be classified as being truly resistant to plum curculio attack by any definition that would have relevance to commercial production and sale of apples.

Greenbug, Schizaphis graminum (Rondani), represents the most important pest insect of sorghum, Sorghum bicolor (L.) Moench, in the Great Plains of the United States. Biotype E is the most widespread and dominant type not only in sorghum and wheat, Triticum aestivum L., fields, but also on many noncultivated grass species. This study was designed to determine sorghum accession PI 550610 resistance to greenbug biotype E, to map the resistance quantitative trait loci (QTLs) by using an established simple sequence repeat (SSR) linkage map and to identify SSR markers closely linked to the major resistance QTLs. In greenhouse screening tests, seedlings of PI 550610 showed strong resistance to the greenbug at a level similar to resistant accession PI 550607. For QTL mapping, one F₂ population containing 277 progeny and one population containing 233 F_2:3 families derived from Westland A line × PI 550610 were used to genotype 132 polymorphic SSR markers and to phenotype seedling resistance to greenbug feeding. Phenotypic evaluation of sorghum seedling damage at 7, 12, 17, and 21 d postinfestation in the F_2:3 families revealed that resistance variation was normally distributed. Single marker analysis indicated 16 SSRs spread over five chromosomes were significant for greenbug resistance. Composite interval and multiple interval mapping procedures indicated that a major QTL resided in the interval of 6.8 cM between SSR markers Xtxp358 and Xtxp289 on SBI-09. The results will be valuable in the development of new greenbug biotype E resistant sorghum cultivars and for the further characterization of major genes by map-based cloning.

Wheat, Triticum aestivum L., with Russian wheat aphid, Diuraphis noxia (Kurdjumov) (Hemiptera: Aphididae) resistance based on the Dn4 gene has been important in managing Russian wheat aphid since 1994. Recently, five biotypes (RWA1–RWA5) of this aphid have been described based on their ability to differentially damage RWA resistance genes in wheat. RWA2, RWA4, and RWA5 are of great concern because they can kill wheat with Dn4 resistance. In 2005, 365 Russian wheat aphid clone colonies were made from collections taken from 98 fields of wheat or barley, Hordeum vulgare L., in Oklahoma, Texas, New Mexico, Colorado, Kansas, Nebraska, and Wyoming to determine their biotypic status. The biotype of each clone was determined through its ability to differentially damage two resistant and two susceptible wheat entries in two phases of screening. The first phase determined the damage responses of Russian wheat aphid wheat entries with resistance genes Dn4, Dn7, and susceptible ‘Custer’ to infestations by each clone to identify RWA1 to RWA4. The second phase used the responses of Custer and ‘Yuma’ wheat to identify RWA1 and RWA5. Only two biotypes, RWA1 and RWA2, were identified in this study. The biotype composition across all collection sites was 27.2% RWA1 and 72.8% RWA2. RWA biotype frequency by state indicated that RWA2 was the predominant biotype and composed 73–95% of the biotype complex in Texas, Oklahoma, Colorado, and Wyoming. Our study indicated that RWA2 is widely distributed and that it has rapidly dominated the biotype complex in wheat and barley within its primary range from Texas to Wyoming. Wheat with the Dn4 resistance gene will have little value in managing RWA in the United States, based on the predominance of RWA2.

Biotypes are infraspecific classifications based on biological rather than morphological characteristics. Cereal aphids are managed primarily by host plant resistance, and they often develop biotypes that injure or kill previously resistant plants. Although molecular genetic variation within aphid biotypes has been well documented, little is known about phenotypic variation, especially virulence or the biotype’s ability to cause injury to cultivars with specific resistance genes. Five clones (single maternal lineages) of Russian wheat aphid, Diuraphis noxia (Kurdjumov) (Homoptera: Aphididae), determined to be injurious to wheat, Triticum aestivum L., with the Dn4 gene, were evaluated on resistant and susceptible wheat and barley, Hordeum vulgare L., for their ability to cause chlorosis, reduction in plant height, and reduction in shoot dry weight. Variation to cause injury on resistant ‘Halt’ wheat, susceptible ‘Jagger’ wheat, and resistant ‘STARS-9301B’ barley was found among the Dn4 virulent clones. One clone caused up to 30.0 and 59.5% more reduction in plant height and shoot dry weight, respectively, on resistant Halt than other clones. It also caused up to 29.9 and 55.5% more reduction in plant height and shoot dry weight, respectively, on susceptible Jagger wheat. Although STARS-9301B barley exhibited an equal resistant response to feeding by all five clones based on chlorosis, two clones caused ≈20% more reduction in plant height and shoot dry weight than three other clones. The most injurious clones on wheat were not the most injurious clones on barley. This is the first report of variation to cause varying degrees of plant damage within an aphid biotype virulent to a single host resistance gene. A single aphid clone may not accurately represent the true virulent nature of a biotype population in the field.

We examined the physiological and biochemical responses of resistant (‘Halt’ and ‘Prairie Red’) and susceptible (‘TAM 107′) wheat, Triticum aestivum L., to injury by the Russian wheat aphid, Diuraphis noxia (Mordvilko). Photosynthetic capacity was evaluated by measuring assimilation/internal CO₂ (A/C_i) curves, chlorophyll fluorescence, chlorophyll, and nonstructural carbohydrate content. Total protein and peroxidase specific activity also were determined. No significant differences were detected in chlorophyll concentration between aphid-infested and control TAM 107 plants. The aphid-infested resistant cultivars had similar or significantly higher chlorophyll concentrations compared with their respective control plants. Measurements over time showed that infested Halt plants had delays in photosynthetic senescence, Prairie Red plants had photosynthetic rate changes that were similar to control plants, and TAM 107 plants displayed accelerated photosynthetic senescence patterns. The photochemical and nonphotochemical quenching coefficients were significantly higher in infested Halt plants compared with their respective control plants on day 3. Infested TAM 107 plants had significantly higher photochemical quenching compared with control plants at all times evaluated, and they had significantly higher nonphotochemical quenching on day 3. Throughout the experiment, infested Prairie Red plants exhibited photochemical and nonphotochemical quenching coefficient values that were not significantly different from control plants. Total protein content was not significantly different between aphid-infested and control plants for all cultivars. Differences between physiological responses of infested susceptible and resistant cultivars, particularly temporal changes in photosynthetic activity, imply that resistant Halt and Prairie Red wheat tolerate some impacts of aphid injury on photosynthetic integrity.

Identifying and using native insect resistance genes is the core of integrated pest management. In this study, 10 experimental corn, Zea mays L., hybrids and 10 inbred lines were screened for resistance to major ear-feeding insects in the southeastern Coastal Plain region of the United States during 2004 and 2005. Ear-feeding insect damage was assessed at harvest by visual damage rating for the corn earworm, Helicoverpa zea (Boddie), and by the percentage of kernels damaged by the maize weevil, Sitophilus zeamais Motschulsky, and stink bugs [combination of Euschistus servus (Say) and southern green stink bug, Nezara viridula (L.)]. Among the eight inbred lines and two control populations examined, C3S1B73-5b was resistant to corn earworm, maize weevil, and stink bugs. In contrast, C3S1B73-4 was resistant to corn earworm and stink bugs, but not to maize weevil. In a similar manner, the corn hybrid S1W*CML343 was resistant to all three ear-feeding insects, whereas hybrid C3S1B73-3*Tx205 was resistant to corn earworm and maize weevil in both growing seasons, but susceptible to stink bugs in 2005. The silk-feeding bioassay showed that corn earworm developed better on corn silk than did fall armyworm. Among all phenotypic traits examined (i.e., corn ear size, husk extension, and husk tightness), only corn ear size was negatively correlated to corn earworm damage in the inbred lines examined, whereas only husk extension (i.e., coverage) was negatively correlated to both corn earworm and maize weevil damage on the experimental hybrids examined. Such information could be used to establish a baseline for developing agronomically elite corn germplasm that confers multiple ear-feeding insect resistance.

Milling wheat, Triticum aestivum L., infested with low densities of internal feeding insects can result in flour containing insect fragments. The Food and Drug Administration (FDA) enforces a standard or defect action level stating that a maximum of 75 insect fragments per 50 g of flour is allowed. However, the relationship between level of infestation and number of resulting fragments is not well documented, and a more rapid method for enumerating insect fragments is needed. We characterized the number of insect fragments produced from milling small lots of wheat spiked with known densities and life stages of Sitophilus oryzae (L.) (Coleoptera: Curculionidae). Insect fragments were enumerated with near-infrared spectroscopy (NIRS), a quick nondestructive procedure, and with the industry standard flotation method. Results showed that an individual small larva, large larva, pupa, or adult produced 0.4, 0.7, 1.5, and 27.0 fragments, respectively. NIRS-predicted counts of ≤51 (from small larvae), ≤53 (from large larvae), ≤43 (from pupae), or 0 (from adults) indicated that there were <75 actual fragments in that sample, because the upper bound of associated 95% inverse prediction confidence intervals was less than the standard; NIRS-predicted counts of ≥98, ≥117, ≥108, or ≥225 fragments (same life stages as above) signaled that these flour samples contained >75 actual fragments. These data suggest that NIRS could be adopted for rapid assessment of insect fragments resulting from relatively low levels of infestation with immature life states, but that it was not accurate enough for enumerating insect fragments, relevant to FDA standards, resulting from adults.

Laboratory bioassays were carried out to evaluate the effect of insect density (10, 30, 60, and 100 adults), wheat quantity (10, 30, 60, and 100 g), and cracked kernel containment (5, 15, 30, and 50%) on the efficacy of diatomaceous earth (DE). Three beetle species, Sitophilus oryzae (L.), Rhyzopertha dominica (F.), and Tribolium confusum Jacquelin du Val, as well as two DE formulations, Insecto and SilicoSec, and one DE enhanced with pyrethrum, PyriSec (all commercially available) were tested. In the first two series of bioassays, the three DE formulations were applied at three dose rates, 500, 1000 and 1,500 ppm. In the third series, the dose rates used were 500 and 1,000 ppm. Dead adults were counted 14 d later. For insect density, wheat quantity, and cracked kernel containment, significant differences were noted in mortality levels of the tested species among the three DE formulations and among doses. No significant differences were noted in the mortality levels among the four adult densities of any of the insects tested. The increase of wheat quantity used in the bioassays increased significantly adult mortality of T. confusum. The increase of cracked wheat containment decreased significantly adult mortality of S. oryzae.

We investigated the ability of two populations of Anisopteromalus calandrae (Hymenoptera: Pteromalidae), to parasitize and develop on late instars of five different stored-product insects that typically complete their development inside seeds of grain or legume species or other dry commodity. The host species were the cigarette beetle, Lasioderma serricorne (F.); cowpea weevil, Callosobruchus maculatus (F.); rice weevil, Sitophilus oryzae (L.); lesser grain borer, Rhyzopertha dominica (F.); and Angoumois grain moth, Sitotroga cerealella (Olivier). Experiments were conducted in the laboratory in a no-choice design by using petri dishes (15 by 100 mm) as experimental arenas with 20 host larvae. A. calandrae females from populations originating in Georgia (GA) and Oklahoma (OK) were introduced singly into experimental arenas and allowed to sting and oviposit for 24 h. Parasitism by the OK population was greater than that for the GA population across all hosts. However, no or very low parasitism was found on Angoumois grain moth for either population in this experiment. The highest number of parasitoid progeny was recorded on cowpea weevil (15.9) followed by rice weevil (11.5) and cigarette beetle (10.8) for the OK population. A similar trend was observed in the GA population. The highest proportion of female progeny was produced on cowpea weevil (73.0%) by the OK population. Conversely, a higher proportion of female progeny was produced on rice weevil (64.6%) by the GA population than produced by the OK population. Parasitoid adults were significantly larger and heavier when they developed on cowpea weevil irrespective of parasitoid population. The possible application of these results for biological control of stored-product insects is discussed.

The titer of chrysanthemum yellows phytoplasma (CYP, “Candidatus Phytoplasma asteris”) in the three vector species Euscelis incisus Kirschbaum, Euscelidius variegatus Kirschbaum, and Macrosteles quadripunctulatus Kirschbaum (Homoptera: Cicadellidae) was measured after controlled acquisition from infected Chrysanthemum carinatum (Schousboe) (daisy) plants. Phytoplasma DNA was quantified in relation to insect DNA (genome units [GU] of phytoplasma DNA per ng of insect DNA) by using a quantitative real-time polymerase chain reaction (PCR) procedure. The increase in phytoplasma titer recorded in hoppers after they were transferred to plants that were nonhosts for CYP provides definitive evidence for phytoplasma multiplication in leafhoppers. CYP multiplication over time in M. quadripunctulatus was much faster than in E. incisus and E. variegatus. CYP titer was also highest in M. quadripunctulatus, and this was reflected in the latent period in the insect. The mean latent period of CYP in M. quadripunctulatus was 18 d versus 30 d in E. variegatus. M. quadripunctulatus was the most efficient vector, giving 100% transmission for single insects compared with 75–82% for E. incisus or E. variegatus, respectively. By sequential transmission, we analyzed the time course of transmission: E. variegatus were persistently infective for life or until shortly before death. Occasionally, leafhoppers failed to maintain continuity of infectivity even after completion of the latent period. PCR analysis of transmitter and nontransmitter E. variegatus adults showed that some nontransmitters were CYP positive, whereas others were CYP negative. These findings suggest that both midgut and salivary gland barriers play a role in transmission efficiency.
