Toxoplasmosis in wild turkeys (Meleagris gallopavo) is of epidemiological interest because turkeys feed from the ground, and detection of infection in turkeys indicates contamination by oocysts in the environment. During the 2018 spring hunting season in Pennsylvania, fresh (unfixed, not frozen) samples were obtained from 20 harvested wild turkeys and tested for Toxoplasma gondii infection. Hearts from all wild turkeys and skeletal muscle from 1 were bioassayed for T. gondii by inoculation in outbred Swiss Webster (SW) and interferon-gamma gene knockout (KO) mice. Antibodies to T. gondii were detected in 1: 5 dilution of neat serum from 5 of 15 wild turkeys and in fluid from the heart of 1 of 4 wild turkeys with the modified agglutination test (MAT); neat serum was not available from 4 wild turkeys. Viable T. gondii was isolated from hearts of 5 wild turkeys, 1 with MAT of 1: 10, 1 with MAT of 1: 5, and 3 seronegative (MAT < 1: 5). Toxoplasma gondii was isolated from both heart and skeletal muscle in the 1 wild turkey that had skeletal muscle submitted. The KO mice inoculated with tissue from all 5 infected wild turkeys died or were euthanatized when ill, 7–21 days post-inoculation (PI). Tachyzoites were detected in lungs of all KO mice, and the T. gondii strains were successfully propagated in cell culture. The SW mice inoculated with tissues of wild turkeys remained asymptomatic, and tissue cysts were seen in the brains of infected mice when euthanatized in good health at 46 days PI; 1 of the 2 SW mice inoculated with the heart of 1 turkey died on day 26, and tachyzoites were detected in its lung. Genetic typing on DNA extracted from culture-derived tachyzoites using the PCR restriction fragment length polymorphism with 10 genetic markers (SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico) revealed that 4 isolates belonged to ToxoDB PCR-RFLP genotype #5 and 1 was genotype #216.

Coccidiosis is a common disease of camels, and camels are important for the economy of Asia and the Arabian Peninsula. Little is known regarding the prevalence of coccidian parasites in camels in Egypt. Fecal samples collected from the rectums of 200 camels at the Cairo slaughterhouse were processed using the sucrose flotation technique. Eimeria species oocysts were found in 38%. Three Eimeria species were identified: Eimeria cameli–like in 31%, Eimeria rajasthani in 18%, and Eimeria dromedarii in 14%. The morphology of E. rajasthani and E. dromedarii oocysts was identical to that in literature. However, there was great variability in size and structure among E. cameli oocysts; oocysts were 70–100 lm long and truncate to ovoid. Four morphotypes (types 1 to 4) were recognized. Types 1 and 2 oocysts had similar truncate ovoid shape and were dark brown, but their shape indices were different. Both types could be easily distinguished from type 3 (elongate ovoid and light brown). All oocysts were enclosed in a transparent outer covering (capsule), although this capsule was barely seen in types 3 and 4. An extension from the capsule situated in front of the micropyle, referred to as polar cap–like structure (PCL), was characteristic for types 1 and 2. The PCL of type 1 resembled the crown, while in type 2 it looked like a small thickening with a smooth top. The PCL was absent in types 3 and 4 oocysts. The latter was found only in a single oocyst. Experimental infections with E. cameli oocysts and molecular studies are needed to determine whether the monotypes described here are different species or strain variations or both.

Malaria infection threatens millions of people worldwide. Sequestering of Plasmodium-infected erythrocytes within the blood vessels of the brain may lead to a more severe form of disease called cerebral malaria (CM), which is difficult to diagnose and treat. Here we used C57BL/6 mice to establish a model of experimental CM (ECM). Comparing the dosage dependence of ECM induction, we found that inoculation with 1×10³ parasitized erythrocytes had higher efficiency at establishing ECM than 1×10⁶ parasitized erythrocytes. However, the percentage of ECM varied in different experimental batches. Infected mice that developed ECM had elevated serum levels of total cholesterol and decreased serum levels of high-density lipoprotein and low-density lipoprotein cholesterol. In addition, ECM mice exhibited liver and kidney dysfunction. ECM induced by low dose inoculation requires additional verification for efficiency. Biochemical analysis of ECM mice revealed characteristic blood lipid levels. These findings provide new clues for the diagnosis and mechanistic probing of CM pathogenesis.

A peculiar bursate nematode, Zygocaulus nagoensis n. gen. and n. sp. (Trichostrongyloidea: Dictyocaulidae: Mertensinematinae), was described from an alien frog, Polypedates leucomystax (Anura: Rhacophoridae), collected on Okinawa Island, Japan. It is related to Mertensinema and Borrellostrongylus, the only hitherto known genera of Mertensinematinae, but is readily distinguished from them by having only 2 pairs of lateral rays and simple distal ends of the dorsal ray branches.

Avian haemosporidian parasites are particularly diverse and widespread. To date, more than 3,000 distinct cytochrome b lineages have been recorded, of which some present extremely wide geographical distributions, even including multiple continents. Whether these isolates represent one or several cryptic species remains unknown. Here we carried out a case study of SISKIN1, a common haemosporidian parasite lineage belonging to the morphologically described species Haemoproteus tartakovskyi. To shed light on its evolutionary origin, we investigated the divergence between SISKIN1 isolates obtained from siskins and redpolls in Europe (Russia and Sweden) and house finches in North America (Mexico). First, we used sequence capture of a small data set (2 Russian isolates and 1 Mexican isolate) to investigate the genetic structure based on the full-length mitochondrial genome and ∼1,000 genes. The mitochondrial genomes of Russian isolates were identical with each other but differed from the Mexican one at 6 positions. The nuclear divergence between Russian and Mexican isolates was on average 2.8%, close to what has been observed between 2 species of malaria parasites that respectively infect humans (Plasmodium falciparum) and gorillas (Plasmodium praefalciparum). Second, we used the expanded data set (15 samples in total) to investigate the genetic structure in 3 genes known to be involved in host invasion. The European isolates were identical across all sequenced genes, whereas the Mexican isolates were highly diverse. The lack of shared alleles between European and Mexican populations suggests that they might have diverged in isolation without gene flow. From the MalAvi database we examined the lineages most similar to the SISKIN1 barcode fragment (part of the cyt b gene) and found that most of them had been recorded in North and South America. This suggests that the lineage SISKIN1 originated in North America and subsequently spread to Europe. Our analyses support that the cyt b gene barcoding region is a useful marker for identification of avian haemosporidian lineages that can classify them into clusters of closely related parasites, but to further investigate species limits and evolutionary history, molecular data from multiple faster-evolving genes are required.

Blood-sucking leeches, some of which are referred to as medicinal leeches, have caught attention not only because of their medical purposes, but also as study organisms to conduct research within fields as diverse as neurobiology, osmoregulation, ecology, and phylogeny. Of particular interest is the question whether hemophagy in leeches is of single origin or evolved independently several times. A key component in the saliva of hematophagous leeches is hirudin, a strong natural inhibitor of thrombin and hence the blood coagulation cascade. Multiple isoforms of hirudin have been described within and among several leech species and genera, often based on sequence data only. The identification of hirudin-like factors (HLFs) illustrated the necessity to underpin such predictions by functional tests. We overexpressed and purified the hirudin of the North American medicinal leech, Macrobdella decora, and proved its thrombin-inhibiting activity. In addition, analysis of the gene structure of both hirudin and some of the decorsins of M. decora clearly indicated conserved exon and intron positions when compared to genes of hirudins and HLFs of Eurasian medicinal leeches. Our data provide evidence for the incorporation of decorsins into the hirudin superfamily and support the concept of a single origin of blood feeding in jawed leeches.

Exotic species can threaten biodiversity by introducing parasites to native hosts. Thus, it is critical to identify if the same parasite species infects both native and exotic hosts. However, developmental- or environmental-induced morphological variation may render species identification ambiguous. Our study reports a range expansion in the southern United States of the pentastome Raillietiella indica from the Mediterranean gecko, Hemidactylus turcicus, as well as a host expansion into the green anole, Anolis carolinensis, in the anole's native range. Species identification was based on sequence data and male spicule shape. In agreement with a study from Australia, we found that much of the morphological variation in hook measurements, the primary diagnostic traits of raillietiellid pentastomes, was due to development. Here, we explicitly link this developmental variation to instar stage by incorporating experimental infection data obtained from the literature. We also show that the various hook traits are themselves highly correlated and, thus, likely not independent. Taking instar stage and correlated hook variables into account, we directly controlled for development on a composite hook size measurement. Using a large sample size from H. turcicus, we did not find any consistent effects of potential factors (host sex, host snout-vent-length, or parasite intensity) that may result in environmental-induced variation in relative hook size (corrected for body length). However, relative male spicule size tended to be negatively correlated with parasite intensity. In contrast, both pentastome body length and relative hook size significantly varied among host species whereas relative male spicule size was not significantly different among host species. Our study independently supports the conclusions that developmental- and host-induced morphological variations need to be accounted for to accurately identify pentastome species.

Larval tapeworms of Echinococcus granulosus have been viewed as the etiological agent for the zoonotic disease cystic echinococcosis, but the species is a complex readily divided into several species and genotypes. Cystic echinococcosis is an important public health issue. Here, the case of liver hydatid cyst in a donkey and molecular characterization of the cyst is presented. The fluid-filled hydatid cyst materials were obtained from the liver of a necropsied donkey. Genomic DNA was extracted and PCR amplification of mitochondrial 12S rRNA gene as well as partial sequencing of mitochondrial cytochrome c oxidase subunit 1 (COI) gene were performed. All cysts were fertile. Traditional polymerase chain reaction (PCR) amplification of 12S rRNA and COI yielded bands (254 and 446 base pairs, respectively) for all 3 cyst samples. However, partial COI gene sequences were identical to those reported for Echinococcus equinus (formerly E. granulosus genotype G4). Thus E. equinus is still transmitting among the equids in Turkey but the mitochondrial 12S rRNA gene primers may not be sufficient for the molecular characterization of members of the E. granulosus species/genotype complex. Molecular diagnosis must be confirmed by partial COI sequence analysis.

Birds harbor a diverse group of haemosporidian parasites that reproduce and develop in the host blood cells, muscle tissue, and various organs, which can cause negative effects on the survival and reproduction of their avian hosts. Characterization of the diversity, distribution, host specificity, prevalence patterns, and phylogenetic relationships of these parasites is critical to the study of avian host–parasite ecology and evolution and for understanding and preventing epidemics in wild bird populations. Here, we tested whether muscle and liver samples collected as part of standard ornithological museum expeditions can be examined to study the diversity and distributions of haemosporidians in the same way as blood collected from individual birds that are typically banded and released. We used a standard molecular diagnostic screening method for mitochondrial DNA (cytochrome b) of the parasites and found that blood, muscle, and liver collected from the same host individual provide similar estimates of prevalence and diversity of haemosporidians from the genera Parahaemoproteus and Leucocytozoon. Although we found higher prevalence for the genus Plasmodium when we screened blood vs. liver and muscle samples, the estimates of the diversity of Plasmodium from different tissue types are not affected at the community level. Given these results, we conclude that for several reasons existing museum genetic resources collections are valuable data sources for the study of haemosporidians. First, ornithological museum collections around the world house tens of thousands of vouchered tissue samples collected from remote regions of the world. Second, the host specimens are vouchered and thus host identification and phenotype are permanently documented in databased archives with a diversity of associated ancillary data. Thus, not only can identifications be confirmed but also a diversity of morphological measurements and data can be measured and accessed for these host specimens in perpetuity.

Gregarine transmission depends upon the environmental encounter rate between viable infective oocysts and suitable hosts. Many factors determine the abundance and distribution of gregarine oocysts in the environment, but the primary factors are oocyst distribution, environmental persistence, and production rate. Prior studies have demonstrated factors affecting oocyst distribution and environmental persistence, but oocyst production rate is poorly understood. This study addresses the effects of gametocyst size on oocyst production. For each of 3 gregarine species, gametocyst size was determined, and the subsequent oocyst production of each gametocyst was quantified. Gregarine species with larger gametocysts produced more oocysts per gametocyst than species with smaller gametocysts. Likewise, within species, larger gametocysts produced more oocysts. The effect was stronger in larger gregarine species, probably as a reflection of the lower overall range of gametocyst size in the smaller gregarine species.

Human infestation with head lice, Pediculus humanus capitis De Geer, is the most prevalent ectoparasitic condition in the modern world. The purpose of this study was to test human head lice from Madagascar for infection with 2 louse-borne bacteria, Bartonella quintana and Acinetobacter spp. including Acinetobacter baumannii, to assess the potential risk of exposure to these pathogens in rural populations experiencing head-louse pediculosis. A second aim was to determine the occurrence of a biomarker for permethrin resistance in head lice from 6 isolated human communities in Madagascar. Deoxyribonucleic acid (DNA) of B. quintana was detected using species-specific Fab3 gene TaqMan in 12.6% of lice from 4 villages. DNA of Acinetobacter spp. was detected using rpoB TaqMan in 42.1% of lice collected from all locations; 58.3% of rpoB-positive lice had the bla_OXA51-like enzyme gene specific for A. baumannii. The kdr-resistant allele was detected in 70% of lice tested and was found in lice from each location. These results provide the first information regarding these combined characteristics of head-louse infestations in Madagascar. This approach can be applied to larger and broader surveys of lice from pediculosis capitis occurring in other geographic locations.

Members of the genus Linognathoides are ectoparasites of ground squirrels and marmots (Rodentia: Sciuridae) in the Nearctic, Palearctic, and Afrotropical regions. Linognathoides urocitelli n. sp. is described based on adult male and female and third-instar nymphal specimens collected from the long-tailed ground squirrel (Urocitellus undulatus) in Mongolia. The new species is compared morphologically to other members of the genus Linognathoides. Additionally, DNA sequences of a 610–base pair (bp) fragment of the nuclear 18S rDNA gene, a 452-bp fragment of the mitochondrial 12S rrnS gene, and a 363-bp fragment of the mitochondrial 16S rrnL gene are provided. Host associations and geographical distributions of the 11 previously recognized species of the genus, and of Linognathoides urocitelli n. sp., are reviewed. A dichotomous identification key to adults of all known species in the genus Linognathoides is provided.

Stool ova and parasite examination using concentration methods remains the gold standard for the investigation of digestive parasitosis. Recently, single-use filtration devices have been marketed for stool concentration sedimentation methods such as MIF or Bailenger, which improve the analytical quality by avoiding contact with feces. In this study, the Parasep^® device was adapted to the Faust technique flotation method. In addition, the performance between conventional techniques (MIF concentration and Faust) and techniques using this device was evaluated on 25 formalin-preserved stools and 3 fresh stools. With the Parasep device, the main parasites (protozoa or helminths) were isolated, and the technical requirements such as hygiene control for the operator and realization according to good laboratory practice were improved due to the filtration device.
