Eighty-eight long-billed dowitchers, Limnodromus scolopaceus, were examined for helminth parasites, 62 from Texas and 26 from Mexico. In total, 3,558 helminth parasites were obtained from this host, 2,273 from Texas birds and 1,285 from birds from Mexico. The component communities consisted of 22 species of helminths in Texas, and 19 in Mexico. Of a total of 26 helminth species recorded from the 2 localities, 15 were common to both, 7 found only in Texas, and 4 only in Mexico. Fifty-nine of 62 Texas birds and 25 of 26 birds from Mexico were infected. The most prevalent helminth for Texas was the cestode Shipleya inermis. The cestode Aploparaksis retroversa was the most abundant, accounting for 37% of the total abundance, and was second highest in prevalence. Five species of cestodes, A. retroversa, Aploparaksis diagonalis, Aploparaksis occidentalis, Aploparaksis rissae, and Shipleya inermis accounted for 79% of total abundance. In the sample from Mexico, S. inermis was also highest in prevalence, followed by the nematode Hystrichis tricolor. The cestode A. retroversa was highest in abundance at 50% of the total, and was third highest in prevalence. Mean species richness, diversity, and evenness were similar among the component communities of Texas and Mexico. A predictable suite of aploparaksid cestodes, together with the cestode S. inermis, constituted 79%, and 61%, of total abundance for the component communities of Texas and Mexico, respectively, and were present in all component communities for locality, season, and year. The cestodes, A. retroversa and S. inermis, were the dominant species in all component communities. Differences among component communities and low similarities for all other comparisons were largely caused by less predictable suites of helminth species. A checklist of helminth parasites reported for long-billed dowitchers is included.

Galaxias maculatus (small puyen) is an abundant native fish distributed in lakes and rivers of the Patagonia, and it is the frequent prey of other fishes, fish-eating birds, and mammals. Previous studies have shown that it is parasitized by 33 metazoan species and that the richness and composition of the parasite communities vary between lakes. The aim of the present work was to analyze the relationship between the composition of fish assemblages and the helminth component community structure of G. maculatus. Ten environmentally similar, small, shallow lakes, belonging to the Nahuel Huapi Lake basin, were chosen because of the differences in the native fish assemblages. Parasite community structure in G. maculatus varied according to the fish assemblage of each lake. The presence of the piscivorous fish Percichthys trucha regularly produced variations in the composition and richness at the component and infracommunity levels, as well as the percentage of autogenic parasite species in G. maculatus.

Twenty-four female ICR mice, 12 acclimated to a 12∶12 light–dark cycle and 12 to a 12∶12 dark–light cycle for 7 days, were each infected with 10 metacercariae of Echinostoma caproni. Infected mice were maintained on their respective lighting regimes for 28 days. Six mice (3 from each group) were necropsied at 4-hr intervals beginning at 0700 hr. The small intestine was removed, opened, and the position of individual worms and worm clusters was measured to the nearest 0.1 cm. Each intestine was subsequently divided into 20 equal segments and individual worms and worm clusters were assigned to the appropriate segment based on the original measurements. All worms were found in the posterior 55% of the intestine (ileum). All posterior segments (10–20), with the exception of segment 18, harbored at least 1 worm at some time. A Monte Carlo simulation of worm abundance in segments 10–17 over all time periods indicated a random distribution, while the same analysis of segments 10–20 indicated a non-random distribution due to large numbers of worms in segment 20 and to the absence of worms in segment 18. To analyze temporal changes in worm distribution, mice were grouped by time of necropsy as follows: night (1900 and 2300 hr), morning (0300 and 0700 hr), and day (1100 and 1500 hr). During the night and morning, E. caproni was heavily concentrated in segments 10–17 and, during the day, worms were located more posteriorly, with a heavy concentration in the last segment (20).

There is limited information about the natural history of the transmission of feather lice (Phthiraptera) from parent birds to their young. This article therefore examines the transmission of 4 species of feather lice from parent blackbirds to their nestlings in an English population, and addresses questions formulated from the perspective of the lice. The lice that disperse onto the several young in the nest were mostly found on the larger chicks, those with higher survival prospects. The lice dispersing to chicks were overwhelmingly nymphs, which cannot be sexed morphologically, and so the prediction that the adult lice dispersing would be disproportionately female, potential founders of a new population, was only supported for the most numerous species, Brueelia merulensis. There was no evidence that louse dispersal to chicks was density dependent and more likely when the parents were more heavily infested. Finally, I predicted that lice might aggregate on female blackbirds, which undertake more brooding, to increase their chance of transmission to nestlings. For 1 louse species, B. merulensis, prevalence, but not louse intensity, was higher on female than male blackbirds. For 2 other louse species, Philopterus turdi and Menacanthus eurysternus, no differences between male and female blackbirds were detected.

Empruthotrema chisholmae n. sp. is described from specimens recovered from a bull ray Pteromylaeus bovinus (Geoffroy Saint-Hilaire, 1817) at the Oceanogràfic Aquarium in Valencia, Spain. The bull ray was caught in the Spanish Mediterranean (Puerto de Mazarrón, Murcia). The new species resembles 4 others of the same genus (Empruthotrema dasyatidis Whittington and Kearn, 1992, Empruthotrema kearni Whittington, 1990, Empruthotrema stenophallus Chisholm and Whittington, 2005, and Empruthotrema tasmaniensis Chisholm and Whittington, 1999) in having a haptor with 13 marginal loculi, the posteriormost loculus single and medial. The new species can be distinguished from these other species of the genus by the morphology of the sclerotized male copulatory organ, which is the shortest described. The new species also differs from the other species by the following combination of features: haptor with 13 marginal loculi, the presence of eyespots, the absence of an accessory piece associated with the male copulatory organ, and a long egg appendage (more than 150 µm). Empruthotrema chisholmae is the first species of the genus reported from the Mediterranean.

We used PCRs with ompA primers to determine if spotted fever group rickettsiae occurred in Amblyomma variegatum from 6 Caribbean islands. Positive amplicons were obtained from ticks from the U.S. Virgin Islands (9/18; 50%), Dominica (39/171; 30%), Montserrat (2/5; 40%), Nevis (17/34; 50%), St. Kitts (46/227; 20%), and St. Lucia (1/14; 7%). Sequences for a convenience sample of reaction products obtained from A. variegatum on St. Kitts (7), American Virgin Islands (4), Montserrat (2), and St. Lucia (1) were 100% homologous with that of Rickettsia africae, the agent of African tick-bite fever. To determine if transmission of R. africae occurred, we used Rickettsia rickettsii antigen in IFA tests and found positive titers (≥1/80) with sera from cattle, goats, and sheep from Dominica (24/95 [25%], 2/136 [2%], 0/58 [0%]), Nevis (12/45 [27%], 5/157 [3%], 0/90 [0%]), St. Kitts (2/43 [5%], 1/25 [4%), 1/35 [3%]), and St. Lucia (6/184 [3%] cattle), respectively. No seropositive animals were found in Grenada (0/4, 0/98/, 0/86), Montserrat (0/12, 0/26, 0/52), or Puerto Rico (0/80 cattle). Our study indicates that R. africae and African tick-bite fever are widespread in the Caribbean.

Nothoaspis amazoniensis n. sp. (Acari: Ixodida: Argasidae) is described from adult and immature ticks (nymph II, nymph I, larva) collected from bat caves in the Brazilian Amazon. Also, 16S rDNA sequences are provided. The diagnostic characters for adults are the presence of false shield or nothoaspis, an anteriorly projecting hood covering the capitulum, a medial extension of palpal article I (flaps), genital plate extending from coxa I to IV, absence of 2 setae on the internal margin of the flaps, a minute hypostome without denticles, presence of a central pore in the base of hypostome, and a reticulate surface pattern on the posterior half of the nothoaspis in males. The nymph II stage is characterized by a hood that is small in relation to the capitulum, short coxal setae, palpal flaps lacking setae on the internal margin, long hypostome, pointed with dentition 4/4 apically, and the anterior half of the body is covered by a cell-like configuration. Nymph I stage is characterized by a hood, small in relation to the capitulum, dorsum of the body covered by a cell-like configuration, venter integument covered by a cell-like configuration, and hypostome dentition 4/4 with apices that are “V”-shaped. Diagnostic characters of the larvae are the number and size of dorsal setae, and the shape of scutum and hypostome. The new species appears to have a life cycle with a larva that feeds on bats, a non-feeding nymphal stage (nymph I), a feeding nymphal stage (nymph II), and adults that probably represent non-feeding stages.

The tegumental ultrastructure of the intestinal fluke Bucephalus anguillae was studied with the use of scanning and transmission electron microscopy. The surface of the tegument is covered by transverse ridges from which protrude numerous closely packed, digitated, and claw-shaped spines. Cobblestone-like units of the tegument were observed on the crescent-shaped formation of the rhynchus and at the posterior part of the body. Three types of sensory structures were examined, i.e., 2 uniciliated receptors and 1 without cilia. As anterior–posterior differences were observed, particular attention was given to spines and sensory receptors. Spine insertion zones and average cilia length are variable between anterior and posterior tegument areas. Ultrastructural study revealed that the tegument of B. anguillae has a typical syncytial organization with a distal cytoplasm lying over a basal matrix and cytons below. Cytoplasmic bridges allowed transit of secretory vesicles and granules. Diagrams of spines and sensory receptors were made to help in understanding the nature of these structures.

Cats are important in the epidemiology of Toxoplasma gondii because they are the only hosts that excrete environmentally resistant oocysts in feces. In the present study, 115 viable T. gondii isolates from tissues of cats from Egypt were genotyped using 10 PCR-restriction fragment length polymorphism markers (SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico) and DNA from tachyzoites. Seven genotypes were recognized including the clonal Type II, Type III (2 genotypes), and 4 atypical genotypes. Ninety percent (103 of 115) of isolates were clonal, i.e., Type II (n  =  61) and Type III (n  =  42) strains. Of the 61 Type II strains, all had the Type II alleles at all loci, except for 2 strains that had allele I at Apico. Eight isolates were divided into 4 atypical genotypes. One of these genotypes (with 4 isolates) was previously reported in dogs from Sri Lanka and in sand cats from the United Arab Emirates. Four isolates had mixed infections. These results revealed a strong clonal population structure with the dominance of clonal Type II and III lineages of T. gondii in feral cats from Egypt.

Cats are important in the epidemiology of Toxoplasma gondii because they are the only hosts that excrete environmentally resistant oocysts in feces. In the present study, 158 feral cats from Giza, Egypt, were examined for T. gondii infection. Antibodies to T. gondii were found in 97.4% with the modified agglutination test. Viable T. gondii was isolated from tissues (brain, heart, tongue) of 115 of 137 cats by bioassay in mice. These isolates were designated TgCatEg 1–115; none of these isolates was virulent to out-bred Swiss Webster mice. Of the 112 seropositive cats whose tissues were bioassayed individually, T. gondii was isolated from the hearts of 83 (74.1%), tongues of 53 (47.3%), and brains of 36 (32.1%). Toxoplasma gondii oocysts were not detected in rectal contents of any of the 158 cats, probably related to high seropositivity (chronic infection) of cats surveyed. The high prevalence of T. gondii in feral cats in Egypt reported here indicates a high environmental contamination with oocysts.

We examined the prevalence of antibodies to zoonotic protozoan parasites (Trypanosoma cruzi, Toxoplasma gondii, and Encephalitozoon cuniculi) and protozoans of veterinary importance (Neospora caninum, Sarcocystis neurona, and Besnoitia darlingi) in a population of North American opossums (Didelphis virginiana) from Louisiana. Samples from 30 opossums were collected as part of a survey for T. cruzi in Louisiana. Frozen sera from these 30 opossums were examined using an indirect immunofluorescent antibody test (IFAT) against in vitro-produced antigenic stages of these protozoans. Additionally, 24 of the 30 samples were examined using hemoculture, and all 30 were examined in the modified direct agglutination test (MAT) for antibodies to To. gondii. The prevalences of reactive IFAT samples were as follows: 60% for T. cruzi, 27% for To. gondii, 23% for E. cuniculi, 17% for S. neurona, 47% for B. darlingi, and 0% for N. caninum. Hemoculture revealed that 16 (67%) of 24 samples were positive for T. cruzi, compared to 18 of 30 (60%) by IFAT. The sensitivity and specificity for the IFAT compared to hemoculture was 100% for each. The modified direct agglutination test revealed that 9 (30%) of the 30 samples from opossums had antibodies to To. gondii, compared to 8 (27%) using the IFAT. The sensitivity and specificity of the IFAT compared to the MAT was 100% and 72%, respectively.

A new species of dicyemid mesozoan is described from Benthoctopus sibiricus Loyning, 1930, collected in the eastern Chukchi Sea. Dicyemennea chukchiense n. sp. is a medium species that reaches about 2,000 µm in length; it lives in folds of the renal appendages. The vermiform stages are characterized in having 23 peripheral cells, a conical calotte, and an axial cell that extends to the base of the propolar cells. An anterior abortive axial cell is present in vermiform embryos. Infusoriform embryos consist of 37 cells; a single nucleus is present in each urn cell and the refringent bodies are solid. This is the first dicyemid described from the Arctic waters.

Baylisascaris procyonis is an intestinal nematode of raccoons (Procyon lotor) that can cause fatal larval migrans in numerous species of birds and mammals, including humans. Although this parasite has historically been absent in the southeastern United States, it has been found in isolated regions in the Appalachian Mountains and was recently documented in DeKalb County, Georgia. The first objective of the current study was to investigate the distribution and prevalence of B. procyonis in selected populations of raccoons in Georgia. Intestinal tracts of 312 raccoons from 25 Georgia counties were examined for B. procyonis. The only county where B. procyonis was detected was Clarke County, where 12 of 116 (10.3%) raccoons were infected. In Clarke County, significantly more juveniles (P  =  0.049) were infected compared with adults, and no differences in prevalence were noted by sex, season of capture, or land use (rural vs. urban); however, significantly (P  =  0.0370) higher worm burdens were found in infected raccoons from urban/suburban locations compared with rural areas. In addition, Toxascaris leonina, a morphologically similar ascarid, was found in 3 raccoons from Clarke County (n  =  2) and Morgan County (n  =  1). A second objective was to determine if sequence polymorphisms were associated with B. procyonis from different geographic regions. Because sequences from a single worm from Japan had been entered into GenBank, we obtained nematodes from Kentucky and Texas for comparison with our samples from Georgia. Sequence analysis of the 18S and 5.8S rRNA genes and the internal transcribed spacer (ITS) -1 and ITS-2 regions confirmed Georgia samples were B. procyonis. Although several polymorphic bases were observed within both ITS regions, none was associated with a particular geographic location. These data indicate that the distribution of B. procyonis within Georgia is increasing and only limited genetic variation is present in the rRNA and ITS gene regions among B. procyonis from the southern United States and introduced populations in Japan.

Leishmania chagasi causes visceral leishmaniasis, a potentially fatal disease of humans. Within the sand fly vector, L. chagasi replicates as promastigotes which undergo complex changes in morphology as they progress from early stage procyclic promastigotes, to intermediate stage leptomonad and nectomonad promastigotes, and ultimately to terminal stage metacyclic promastigotes that are highly infective to vertebrates. This developmental progression is largely recapitulated in vitro using axenic promastigote cultures that have been passaged only a few times. Within a single passage (which takes about a week), axenic cultures progress from logarithmic to stationary growth phases; parasites within those growth phases progress from stages that do not have metacyclic cell properties to ones that do. Interestingly, repeated serial passage of promastigote cultures will result in cell populations that exhibit perturbations in developmental progression, in expression levels of surface macromolecules (major surface protease, MSP, and promastigote surface antigen, PSA), and in virulence properties, including resistance to serum lysis. Experiments were performed to determine whether there exists a direct relationship between promastigote developmental form and perturbations associated with repeated serial passage. Passage 2 to passage 4 L. chagasi cultures at stationary growth phase were predominately (>85%) comprised of metacyclic promastigotes and exhibited high resistance to serum lysis and high levels of MSP and PSA. Serial passaging 8, or more, times resulted in a stationary phase population that was largely (>85%) comprised of nectomonad promastigotes, almost completely devoid (<2%) of metacyclic promastigotes, and that exhibited low resistance to serum lysis and low levels of MSP and PSA. The study suggests that the loss of particular cell properties seen in cells from serially passaged cultures is principally due to a dramatic reduction in the proportion of metacyclic promastigotes. Additionally, the study suggests that serially passaged cultures may be a highly enriched source of nectomonad-stage promastigotes, a stage that has largely been characterized only in mixtures containing other promastigote forms.

Balantidium coli is a ciliate reported in many mammalian species, including African great apes. In the former, asymptomatic infections as well as clinical balantidiasis have been reported in captivity. We carried out a cross-sectional study of B. coli in African great apes (chimpanzees, bonobos, and both species of gorillas) and examined 1,161 fecal samples from 28 captive facilities in Europe, plus 2 sanctuaries and 11 wild sites in Africa. Samples were analyzed with the use of Sheather's flotation and merthiolate–iodine–formaldehyde (MIFC) sedimentation. MIFC sedimentation was the more sensitive technique for diagnostics of B. coli in apes. Although not detected in any wild-ape populations, B. coli was diagnosed in 52.6% of captive individuals. Surprisingly, in the apes' feces, trophozoites of B. coli were commonly detected, in contrast with other animals, e.g., Old World monkeys, pigs, etc. Most likely reservoirs for B. coli in captive apes include synantropic rats. High starch diets in captive apes are likely to exacerbate the occurrence of balantidiasis in captive apes.

We characterized the complete nucleic and amino acid sequences of the Plasmodium inui circumsporozoite protein (Pincsp) gene and analyzed nucleotide diversity across the entire Pincsp gene by using 7 field isolates and strains Taiwan I and II obtained from Formosan macaques (Macaca cyclopis) in Taiwan. The length of the circumsporozoite protein (CSP) gene ranged from 1077 to 1125 bp. Size polymorphisms were due to variations in the number of tandem repeat units. The non-repetitive (NR) region exhibited high homology (99.1∼100 and 98.7∼100% at the nucleotide and amino acid levels, respectively) and was conserved among the variants (nucleotide diversities, π, of the 5′NR and 3′NR regions were 0.00364 and 0.00392, respectively). In the central repetitive (CR) region, we decomposed the sequences into 2 kinds of repeating amino acid motifs, i.e., a repeat unit R1, PA(P/A)(P/A)A(E)GG (n  =  11–13), and a following repeat unit R2: P(A/G)(A/P/G)(P/Q)AQ(N/K) (n  =  9–10). Analyzing these repeat sequences showed evidence of 3 genetic mechanisms for generating variations in the repeats of the Pincsp gene, i.e., point mutation, insertion, and recombination. These findings suggest that polymorphisms in the Pincsp gene are essentially limited to the CR region, which showed much greater variability in terms of length, number of repeats, and sequence.

Dioctophyma renale is a parasite that frequently occurs in animals but rarely in humans. The present report describes the clinical observations of a D. renale infection in a 51-yr-old woman. Its clinical signs and diagnostic findings were unspecific until giant worms were observed in the urine and histological findings confirmed it was a D. renale infection. She refused treatment and died of bilateral renal function failure. This is the first confirmed report to follow the natural progression of D. renale infection in a human. Here, we discuss a conservative therapeutic approach and features associated with this parasitic infection.

A new species of a microsporidium found in the freshwater teleost Gymnorhamphichthys rondoni, collected on the lower Amazon River, is described based on light, ultrastructural, and phylogenetic studies. This parasite develops in the skeletal muscle of the abdominal cavity, forming whitish cyst-like structures containing numerous spores. Mature spores, lightly pyriform to ellipsoidal with rounded ends and measuring 4.25 ± 0.38 × 2.37 ± 0.42 µm (n  =  30), were observed. The spore wall, which measured about 102 nm, was composed of 2 layers with approximately the same thickness. The isofilar polar filament was coiled, with 9–10 (rarely 8) turns. The posterior vacuole appeared as a pale area, occupying about 1/3 of the spore length, and contained a spherical posterosome composed of granular material that was denser at the periphery. The myofibrils located near the spores appeared to be in advanced degradation. Molecular analysis of the rRNA genes, including the ITS region, and phylogenetic analyses using maximum parsimony, maximum likelihood, and Baysesian inference were performed. The ultrastructural characteristics of the spores and the phylogenetic data strongly suggested that it is a new species related to Kabatana, Microgemma, Potaspora, Spraguea, and Tetramicra. We named this new microsporidian from Amazonian fauna as Kabatana rondoni n. sp.

Myxobolus burti n. sp. is described from striated muscle of Notropis hudsonius (Cyprinidae) collected from localities in Lake Superior, Lake Michigan, Lake Huron, Lake St. Claire, Lake Erie, Lake Ontario, and the St. Lawrence River. Myxobolus burti is intracellular, forming thin-walled, cigar-shaped plasmodia within striated muscle cells of the body. It exhibits disporoblastic, oval spores, approximately 10 µm long, 8 µm wide, and 6 µm thick, with a length-to-width ratio of 1.3∶1. They resemble most closely those of Myxobolus bellus Kudo, 1934, and Myxobolus mutabilis Kudo, 1934, but have polar capsules unequal in length (5.3 vs. 4.7 µm) and oblique filament coils. The new parasite was not encountered in routine examination of other small-bodied fishes at collection locations and thus looks to be specific to N. hudsonius. A comparative plate of similar species encountered during surveys of these fishes in the northeast is presented to contrast the uniqueness of the new species.

Terrestrial tortoises, Chersina angulata (Schweigger, 1812), Kinixys belliana belliana Gray, 1831, K. lobatsiana Power, 1927, K. natalensis Hewitt, 1935, and Stigmochelys pardalis (Bell, 1828), both wild (n  =  84) and captive (n  =  70), were examined for hematozoans from the provinces of Gauteng, Kwazulu-Natal, North West, and Western Cape, South Africa. In Giemsa-stained blood films, 2 species of hemoproteids were identified, 1 previously described, that is, Haemoproteus testudinalis (Laveran, 1905) Wenyon, 1915, the other unknown. Haemoproteus testudinalis occurred in 2/27 wild K. lobatsiana and 1/41 S. pardalis and 2/2 captive K. lobatsiana from the North West and Gauteng, respectively, whereas Haemoproteus sp. A was recorded in 1 of 3 captive K. natalensis from Kwazulu-Natal. Haemoproteus testudinalis had pale blue-stained, often vacuolated, halteridial macrogametocytes, pale pink-stained microhalterdial to halteridial microgametocytes, and dark, scattered, often bacillus-like hemozoin granules. Haemoproteus natalensis n. sp. had ameboid growth stages, blue-stained halteridial to circumnuclear macrogametocytes, purple-stained circumnuclear microgametocytes, and brownish, often clustered, pigment granules. It is recommended that Haemoproteus balazuci Dias, 1953, become a junior synonym of H. testudinalis.

Hydromermis biesboeri n. sp. (Nematoda: Mermithidae) is described from the hemocoel of imagos of chironomid host species eclosing from Lake Ozawindib in northern Minnesota in July and August 2008 and 2009. The new species is distinguished from the other 28 described species in the genus by terminal mouth, wide oval (male) and long oval (female) amphids with a dorsal commissure, tapered and rounded posterior ends, split base of the spicule, short protractor muscles in males, and by females having an S-shaped vagina with equal-length terminal limbs. The new species represents the 10th Hydromermis mermithid species described, or reported, from the region and the second from Lake Ozawindib.

New collections of cestodes from the spiral intestines of the lanternsharks Etmopterus spinax and Etmopterus pusillus off the island of Faial, in the Azores, Atlantic Ocean, have yielded 2 new species of trypanorhynchs belonging to Aporhynchus. Both species share the distinctive lack of all elements of the rhyncheal system that are characteristic of this genus. The identity of Aporhynchus norvegicus is clarified to allow it to be distinguished from A. menezesi n. sp., which also parasitizes E. spinax. This new species differs conspicuously from its congeners in that its mature and gravid proglottids are wider than long, rather than longer than wide, and also in its lack of spinitriches on the scolex. Aporhynchus pickeringae n. sp., the new species from E. pusillus, differs from all of its congeners except A. norvegicus in that it is a relatively delicate worm with relatively fewer testes. It also possesses fewer proglottids and a wider pedunculus scolecis than does A. norvegicus. Sections through the scolex of A. menezesi n. sp. support use of the term bothriate, rather than difossate, in reference to the scolex configuration of some trypanorhynchs. A key to the 4 species of Aporhynchus is provided.

New collections of cestodes from the spiral intestines of catsharks (Carcharhiniformes: Scyliorhinidae) off Taiwan have led to the discovery of a new genus and 2 new species of trypanorhynchs. These taxa are relatively unique among trypanorhynchs in their lack of all elements of the rhyncheal apparatus. The new genus, Nakayacestus n. gen., is considered to belong with Aporhynchus in the Aporhynchidae. In addition to lacking the rhyncheal apparatus, these 2 genera share circum-medullary vitelline follicles, post-ovarian testes, and complex terminal genitalia consisting of accessory, external, and internal seminal vesicles. The 2 genera differ conspicuously in spinithrix configuration; whereas both species of Nakayacestus n. gen. bear scolex spinitriches that are bifid, trifid, or pectinate, species of Aporhynchus either lack scolex spinitriches entirely or possess spathulate spinitriches. The configuration of the bothria of the 2 genera also differ conspicuously. Whereas the bothria of Aporhynchus are sessile and generally do not extend beyond the lateral margins of the cephalic peduncle, those of Nakayacestus bear only a tenuous connection with the scolex proper, being conspicuously free both anteriorly and posteriorly and extending conspicuously beyond the cephalic peduncle. Futhermore, the boundary between the scolex and the strobila of members of the new genus is clearly delineated, whereas this distinction is ill-defined in species of Aporhynchus. Nakayacestus takahashii n. sp., the type of the new genus, was collected from the Broadmouth catshark, Apristurus macrostomus, and differs from Nakayacestus tanyderus n. sp., collected from the Blacktip sawtail catshark, Galeus sauteri, in being shorter, bearing a longer pedunculus scolecis, an ovary that is more posterior in the proglottid, and fewer post-ovarian testes. Furthermore, the 2 new species differ conspicuously from one another in the configuration of their scolex spinitriches.

Cyclobulura superinae n. sp. collected from Zaedyus pichiy and Chaetophractus vellerosus is herein described as the second species in Cyclobulura Quentin, 1977, and the first subulurid in armadillos. The species is unique in the spur-like structures present at the tip of both spicules, yet they conform to the description of Cyclobulura in the structure of the buccal parts. Specimens of the new species show longer chordal lobes and more conspicuous radial lobes and are smaller than specimens of C. lainsoni. In addition, males of C. superinae exhibit a spur-like process in the distal end of the spicules and a shorter tail (170 vs. 300 µm) with no spine. Finally, the eggs of C. superinae are smaller (60–89 × 45–71 vs. 95–100 × 80–85). To our knowledge, the new species is the first subulurid nematode found in an armadillo.

Inferences about the evolution of host-parasitic relationships are often made based on the prevalence of avian malaria, which is usually estimated in a large sample of birds using either microscopic or molecular screening of blood samples. However, different techniques often have variable accuracy; thus, screening methodology can raise issues about statistical bias if method sensitivity varies systematically across parasites or hosts. To examine this possibility, published information was collected on the prevalence of species in 4 genera of avian blood parasites (Plasmodium, Haemoproteus, Leucocytozoon, and Trypanosoma) from various sources that used different tools. The data were tested to determine if the application of different methods provided different estimates for the same hosts. In these comparisons between the main methodologies, the PCR-based molecular methods were generally found to provide higher estimates for Plasmodium spp. prevalence than microscopic tools, while there was no significant tendency for such a trend in species of Haemoproteus and Leucocytozoon. When analyzing intraspecific variance of prevalence within molecular studies, some studies provided consistently higher estimates for Haemoproteus spp. prevalence than others, indicating that differences between studies can affect detected estimates. Within microscopic studies, surveys that examined more microscopic fields were more likely to report higher prevalence for Plasmodium spp. than those relying on fewer microscopic fields. Consequently, studies making comparisons across parasite genera and/or host species from different sources need to consider several types of bias originating from variation in method sensitivity.

Enhanced plasma availability of albendazole sulphoxide (ABZSO), the active metabolite of albendazole (ABZ), has been described in feed-restricted sheep. The aim of the present work was to determine if the absorption-related pharmacokinetic changes derived from fasting animals prior to drug treatment would modify the clinical efficacy of ABZ against resistant gastrointestinal nematodes in lambs. Forty Corriedale lambs, naturally infected with resistant gastrointestinal nematodes, were divided into 4 groups. Controls were fed ad libitum and did not receive any drug treatment. Treated animals were fed ad libitum up to 30 min prior to treatment with ABZ (3.8 mg/kg) by the intraruminal route. The control (fasted) animals were not fed during the 24-hr period prior to the start of the experiment and did not receive any drug treatment. A second treated group of animals were fasted 24 hr prior to the treatment with ABZ, as previously described for the fed-treated group. Blood samples were collected over a period of 72 hr post-treatment from 6 animals in each treated group. Plasma samples were analyzed by high performance liquid chromatography. The pharmacokinetic parameters were statistically compared using parametric statistical tests. The estimation of the efficacy of the different treatments was performed by the fecal egg count reduction test (FECRT). Additionally, 4 animals randomly chosen from the control-fed and treated groups were killed 13 days post-treatment to evaluate the efficacy against different adult nematode parasites. The results were statistically compared by parametric and non-parametric tests. Significantly (P < 0.05) higher Cmax and AUC values were observed for both the ABZSO and ABZ-sulphone (ABZSO₂) metabolites in the fasted compared to the fed animals. These kinetic results may be due to a fasting-induced delay in the GI transit time which increases ABZ dissolution and GI absorption. However, a poor ABZ efficacy (measured as FECRT), compatible with a high degree of nematode resistance, was obtained in both fed (48%) and fasted (49%) animals. Haemonchus contortus and Trichostrongylus colubriformis appeared as the more reluctant species with respect to ABZ treatment. The efficacy against H. contortus ranged between 37 (fed) and 54% (fasted) and against T. colubriformis between 0% (fed) and 16% (fasted). Under these experimental conditions, the fasting-induced improvement on ABZ systemic availability (>60%) did not improve its activity against nematodes with a high degree of resistance. However, the data described here for a highly resistant nematode population should not discourage the use of fasting as a practical and well-proven management tool for parasite control in ruminants.

A variety of bioactive proteins from medicinal leeches, like species of Hirudo, have been characterized and evaluated for their potential therapeutic biomedical properties. However, there has not previously been a comprehensive attempt to fully characterize the salivary transcriptome of a medicinal leech that would allow a clearer understanding of the suite of polypeptides employed by these sanguivorous annelids and provide insights regarding their evolutionary origins. An Expressed Sequence Tag (EST) library-based analysis of the salivary transcriptome of the North American medicinal leech, Macrobdella decora, reveals a complex cocktail of anticoagulants and other bioactive secreted proteins not previously known to exist in a single leech. Transcripts were identified that correspond to each of saratin, bdellin, destabilase, hirudin, decorsin, endoglucoronidase, antistatin, and eglin, as well as to other previously uncharacterized predicted serine protease inhibitors, lectoxin-like c-type lectins, ficolin, disintegrins and histidine-rich proteins. This work provides a lens into the richness of bioactive polypeptides that are associated with sanguivory. In the context of a well-characterized molecular phylogeny of leeches, the results allow for preliminary evaluation of the relative evolutionary origins and historical conservation of leech salivary components. The goal of identifying evolutionarily significant residues associated with biomedically significant phenomena implies continued insights from a broader sampling of blood-feeding leech salivary transcriptomes.

The seroprevalence of Toxoplasma gondii infection in Tibetan sheep was surveyed in Qinghai Province, northwestern China, in May and June 2010. In total, 580 serum samples was collected from 6 counties, and antibodies to T. gondii were detected by indirect hemagglutination (IHA) test. Antibodies to T. gondii were found in 29.8% (173/580); regional differences showed the highest prevalences in Nangqian County (39.4%) and Zaduo County (48.7%). The results of the present study indicate that infection with T. gondii in sheep is widely spread in China, including Tibetan sheep in Qinghai Province, and is a potential public health concern.

We developed a novel and simple method, using loop-mediated isothermal amplification (LAMP), for the detection and discrimination of Toxocara canis and Toxocara cati eggs. The new method employs 4 steps: (1) concentration of Toxocara eggs in a small amount of sand; (2) dissolution of the proteinaceous membrane of eggs and simultaneously separation of them from the sand using NaClO treatment; (3) extraction of DNA using NaOH treatment; and (4) detection of T. canis/T. cati DNA using a LAMP assay. All these steps are fast, easy to perform, and do not require expensive equipment or reagents. The novel method was tested both experimentally and in a field study. In the laboratory, we could reliably detect as few as 3 T. canis eggs in artificially contaminated sand, if the experiment was repeated twice. In the field trial, we were able to detect T. cati DNA from 4 natural sandpits having moderate to heavy contamination, although not in a single lightly contaminated sandpit. All of the examined sandpits were found to be contaminated with eggs of T. cati, but none appeared to contain T. canis. Our new method could extract DNA from T. canis and T. cati eggs directly from sand samples as well as detect and distinguish these 2 species in a few easy steps, with markedly reduced time and expense.

Filarial thiordoxin peroxidase is a major antioxidant that plays a crucial role in parasite survival. Although Brugia malayi TPx has been shown to be a potential vaccine candidate, it shares 63% homology with its mammalian counterpart, limiting its use as a vaccine or drug target. In silico analysis of TPx sequence revealed a linear B epitope in the host's nonhomologous region. The peptide sequence (TPx peptide_27–48) was synthesized, and its reactivity with clinical sera from an endemic region was analyzed. The peptide showed significantly high reactivity (P < 0.05) against the sera of putatively immune individuals compared to the nonendemic control sera. It also showed high reactivity against the sera of patients with chronic pathology and patent infection. The high reactivity of the peptide with endemic immune sera equivalent to that of whole protein shows that it forms a dominant B epitope of TPx protein and thus could be utilized for incorporation into a multiepitope vaccine construct for filariasis.

American trypanosomiasis and leishmaniasis are caused by related hemoflagellate parasites, Trypanosoma cruzi and Leishmania spp., which share several common host species. Both zoonotic protozoans are endemic in the United States. Canines, including domestic and wild canids, are reservoir hosts for human infections with T. cruzi and Leishmania spp. The present study examined the seroprevalence of T. cruzi and Leishmania spp. in wild canids from North Carolina and Virginia. Wild canine species tested in this work included 49 gray foxes (Urocyon cinereoargenteus) and 5 red foxes (Vulpes vulpes). Overall, sera samples from 54 foxes (North Carolina  =  43; Virginia  =  11) were tested by immunochromatographic strip assays (ICT). Antibodies to T. cruzi were found in 4 (9%) gray foxes from North Carolina and 2 (18%) gray foxes from Virginia. Antibodies to Leishmania spp. were detected in 1 (2%) gray fox from North Carolina. Our results indicate that wild canids are exposed more frequently to T. cruzi in North Carolina than Leishmania spp. and only T. cruzi in Virginia.

In the Peruvian Amazon, the white-lipped peccary (Tayassu pecari) is a desirable game species and is important for the local rural economy. Blood samples from 101 white-lipped peccaries from Peru were collected from 3 different conservation areas located in the municipalities of Manu and Tambopata, southeastern region of the Peruvian Amazon. Antibodies were assayed using the modified agglutination test (MAT, cut of value of 25). Antibodies to Toxoplasma gondii were found in 89.1% (90 of 101) of animals, with titers of 1∶25 in 9, 1∶50 in 25, 1∶100 in 20, 1∶200 in 14, 1∶400 in 12, 1∶800 in 9, and 1∶3,200 in 1; 87.7% and 89.2% of males and females, respectively, tested positively, and no association (P ≥ 0.05) with gender and occurrence of antibodies was observed.

Toxoplasma gondii is widely distributed in humans and other animals, including wild rats throughout the world, but little is known of the prevalence of T. gondii in rats in China. The seroprevalence of T. gondii in rats (Rattus norvegicus and Rattus flavipectus) was investigated in Guangzhou, southern China, between November 2009 and January 2010. In total, 217 rat serum samples were collected; antibodies to T. gondii were detected by the modified agglutination test (MAT), and 7 (3.2%) were found positive (titers ≥1:40). The seroprevalence was higher (3.4%) in R. norvegicus than in R. flavipectus (3.0%), but the difference was not statistically significant (P > 0.05). All 7 positive rats were female; no T. gondii antibodies were detected in males. This is the first extensive survey of T. gondii infection in rats in southern China, and the results have public health implications in this region.
