Before the availability of digital photography resulting from the invention of charged couple devices in 1969, the measurement of plant architecture was a manual process either on the plant itself or on traditional photographs. The introduction of cheap digital imaging devices for the consumer market enabled the wide use of digital images to capture the shape of plant networks such as roots, tree crowns, or leaf venation. Plant networks contain geometric traits that can establish links to genetic or physiological characteristics, support plant breeding efforts, drive evolutionary studies, or serve as input to plant growth simulations. Typically, traits are encoded in shape descriptors that are computed from imaging data. Skeletons are one class of shape descriptors that are used to describe the hierarchies and extent of branching and looping plant networks. While the mathematical understanding of skeletons is well developed, their application within the plant sciences remains challenging because the quality of the measurement depends partly on the interpretation of the skeleton. This article is meant to bridge the skeletonization literature in the plant sciences and related technical fields by discussing best practices for deriving diameters and approximating branching hierarchies in a plant network.

The reticulate venation that is characteristic of a dicot leaf has excited interest from systematists for more than a century, and from physiological and developmental botanists for decades. The tools of digital image acquisition and computer image analysis, however, are only now approaching the sophistication needed to quantify aspects of the venation network found in real leaves quickly, easily, accurately, and reliably enough to produce biologically meaningful data. In this paper, we examine 120 leaves distributed across vascular plants (representing 118 genera and 80 families) using two approaches: a semiquantitative scoring system called “leaf ranking,” devised by the late Leo Hickey, and an automated image-analysis protocol. In the process of comparing these approaches, we review some methodological issues that arise in trying to quantify a vein network, and discuss the strengths and weaknesses of automatic data collection and human pattern recognition. We conclude that subjective leaf rank provides a relatively consistent, semiquantitative measure of areole size among other variables; that modal areole size is generally consistent across large sections of a leaf lamina; and that both approaches—semiquantitative, subjective scoring; and fully quantitative, automated measurement—have appropriate places in the study of leaf venation.

Recent advances in microscopy, imaging, and data analyses have permitted both the greater application of quantitative methods and the collection of large data sets that can be used to investigate plant morphology. This special issue, the first for Applications in Plant Sciences, presents a collection of papers highlighting recent methods in the quantitative study of plant form. These emerging biometric and bioinformatic approaches to plant sciences are critical for better understanding how morphology relates to ecology, physiology, genotype, and evolutionary and phylogenetic history. From microscopic pollen grains and charcoal particles, to macroscopic leaves and whole root systems, the methods presented include automated classification and identification, geometric morphometrics, and skeleton networks, as well as tests of the limits of human assessment. All demonstrate a clear need for these computational and morphometric approaches in order to increase the consistency, objectivity, and throughput of plant morphological studies.
