Litobothrium aenigmaticum Caira, Jensen, Waeschenbach, & Littlewood 2014 is a cestode species that parasitizes pelagic thresher sharks in Taiwan and the Gulf of California. A previous study using light microscopy suggested the scolex contains four types of unusual tissues, one of which was considered to aid in adhesion of the worm to the mucosa of the spiral intestine. The function of the other tissues was unknown. Also unknown was the function of two laterally paired ducts found extending throughout the length of the worm. The goal of the present study was to use transmission electron microscopy (TEM) to examine the internal anatomy of the scolex and cephalic peduncle of this tapeworm in more detail. Three specimens from Taiwan were examined with TEM and two with light microscopy. Transmission electron microscopy revealed that the laterally paired ducts were excretory, since they were closely associated with numerous protonephridia. The number of cell types, previously referred to as tissues, in the scolex was expanded to 11 based on a combination of features including cell size, nucleus size, ratio of the maximum diameter of the nucleus to maximum diameter of the whole cell, and types of observed organelles, as well as the presence or absence of cytoplasmic electron-dense and electron-lucent vesicles. The cytoplasm of all 11 cell types was found to include electron-dense vesicles, and all types were periodic acid-Schiff (PAS) positive. This suggests that the electron-dense vesicles may contain glycoproteins and/or mucoproteins. As all 11 cell types occurred within the region of the scolex that is surrounded by a papilla of host mucosa at the site of tapeworm attachment, we hypothesize that one or more of these cell types may produce a substance that induces this inflammatory reaction. The specific products in the secretory vesicles and how those products are released are still unclear, as are any relationships between or among the cell types in terms of products produced. Further research will be necessary to fully understand the complexity of this organism.

The Humboldt Current krill, Euphausia mucronata (Crustacea: Euphausiacea), is an endemic and keystone species in the food web of this highly productive eastern border current ecosystem. The morphology and ontogeny of E. mucronata is known from calyptopis I to the adult phase, but the embryonic and early-life stages (nauplius and metanauplius) of this broadcast spawning species are unknown. We describe the morphology and development time of these life stages to complete the knowledge of its life cycle. Embryos were obtained from purple-gonad gravid females collected off Dichato, central Chile, during November 2012. Eight gravid females (mean=16 mm total length) were incubated in seawater at 12°C under laboratory conditions. The average development time from single-cell embryos to the metanauplius stage was 2.2 d and hatching occurred between 20 and 25 h. The average growth rate was 0.35 mm d^–1 from the late limb bud to the metanauplius stage (range=0.21–0.48 mm d^–1). Embryos of E. mucronata had a mean chorion diameter of 0.460 mm, embryo diameter of 0.343 mm, and perivitelline space of 0.056 mm. Our biological information reported here constitutes a baseline for future ecological studies on distribution and temporal variability of spawning activity and reproductive strategies of E. mucronata in the highly variable and productive Humboldt Current ecosystem.

The reproductive systems of crabs reveal characters of considerable importance for the understanding of brachyuran phylogeny and evolution. The Dorippoidea show several plesiomorphic characters within Eubrachyura and similarities to podotreme crabs. Hence, they are often considered as an early diverging lineage, sometimes even as the sister group to all remaining eubrachyurans. Due to their role as prime candidates for putative plesiomorphic characters of the reproductive system of the Eubrachyura, we compared the morphology of the vaginae, seminal receptacles, and ovaries of three dorippid species using histological methods, micro-computed tomography, and 3-D reconstructions. Despite the putative phylogenetic position of dorippids, the female reproductive system shows features that are regarded as derived characters in eubrachyurans, including a concave vagina and a ventral-type seminal receptacle. In contrast to other eubrachyurans, the oviduct does not enter the seminal receptacle directly but through specific cuticular valves. The female reproductive systems of Dorippe sinica and Dorippe quadridens are remarkable in further aspects. The seminal receptacles of both species are completely cuticle-lined and have accessory sperm storage structures, the bursae. Our findings on the morphology of the female reproductive system of dorippids with its unique combination of basal, derived, and new characters challenges the prevailing hypothesis on the evolution of sperm storage organs in Eubrachyura.

In the pteropod mollusc Clione limacina Phipps 1774, individuals possess an open circulatory system that fills their body cavities and functions as a hydrostatic skeleton. Individuals of C. limacina demonstrate two distinct swimming behaviors, slow and fast swimming, and their wings are supported by their hydrostatic skeleton. We investigated the circulation of fluid within the body cavities of individuals of C. limacina by injecting dye into the hemocoelic compartments to visualize flow during both slow swimming and serotonin-induced fast swimming. Hemocoelic fluid was observed to have a defined pattern of flow: rostrally from the heart into the wings and head, then following a dorsal pathway caudally into the body and tail before being taken up by the heart again. During patterned attack behavior, the neck constricted in width as the head's buccal cones were hydraulically inflated with hemocoelic fluid.

Differential loss of heterozygous individuals that move to the periphery of mussel aggregations, where they are at greater risk for dislodgement, has been proposed as an explanation for observed heterozygote deficiencies in blue mussels. To test the dislodgement hypothesis, correlations between heterozygosity and mussel motility, as well as characteristics of byssogenesis and byssal thread attachment strengths, were determined in a wild and a farmed population of blue mussels (Mytilus edulis) from New Hampshire, USA. Although both populations exhibited a heterozygote deficit as measured by three microsatellite loci, no relationship was found between heterozygosity and increased motility in either population. Similarly, no relationship was found between heterozygosity and byssogenesis or attachment strength. Hence, differential dislodgement is highly unlikely as a possible contributor to the loss of heterozygous individuals.

The invasive kudzu bug, Megacopta cribraria, was first reported in North America in 2009 and has subsequently spread through most of the southeastern United States, causing yield loss in soybean. Since detection in the USA, research has focused mainly on managing this newly established pest, but many important characteristics of the pest's mouthpart morphology and feeding behavior are unknown. Qualitative and quantitative comparisons of nymph and adult mouthparts and sensilla were made through scanning electron microscopy and light microscopy, and feeding behavior was examined using electropenetrography (EPG) and paraffin histology. Morphologies observed were similar to what has previously been reported for other piercing–sucking hemipterans. The relationship between rostrum length and body size (pronotum width and dorsal length) exhibited negative allometry. Rostrum length exhibited an isometric relationship with interocular width. Adult females (n=9) probed soybean stems 1.3±0.8 times in 9 h, with an average probe time of 2.3±1.3 h. EPG waveforms were characterized and correlated with behavior. Salivary sheaths were shown to terminate in the vascular tissue; four of five sheaths terminated in the phloem. This is the first time that the feeding behavior of a member of the Plataspidae has been recorded using EPG. Results add to our current limited knowledge of plataspid mouthpart morphology and provide a baseline for further research on the feeding behaviors of M. cribraria and other soybean-feeding hemipterans.

The main difference between the sponge genera Leucilla and Paraleucilla (Porifera, Calcarea, Amphoriscidae) is the presence of a disorganized zone (DZ) in the inner region of the skeleton of the latter genus. However, it has been repeatedly observed that specimens from different species of Paraleucilla lack this feature. It is assumed that the size of the sponge may have an effect on the presence or the absence of the DZ, but no investigation of this morphological variation has previously tested this hypothesis. Here, we examined this assumption and described the frequency with which the DZ is absent from individuals of Paraleucilla magna. We also investigated possible drivers of the observed variation using generalized linear models to evaluate whether the month of the year, rainfall, reproductive activity, volume, and body wall thickness could influence the presence or the absence of the DZ. The DZ was absent from 46.5% of the analyzed individuals, indicating that it may be misleading to use this trait to identify the genera Leucilla and Paraleucilla. The presence of the DZ in P. magna is influenced negatively by volume and positively by body wall thickness of the individuals. Our results confirm the previous assumptions for the family Amphoriscidae and highlight problems with the current classification of Calcarea. A discussion of the validity of some morphological characters and the importance of analyzing their variation is provided.

Energy-converting symbionts and organelles may be central to the cnidarian stress response. Stress may damage photochemistry in the endosymbiotic dinoflagellate Symbiodinium spp., leaving photosystems I and II relatively reduced and increasing reactive oxygen species (ROS). Alternatively, ROS may emanate from mitochondria of either the host or the symbiont, or both. These sources of ROS can be distinguished by using microscopy to examine the effects of light on stressed cnidarians incubated in the dark with a fluorescent, ROS-detecting probe. Experiments were carried out with three species of alcyonacean octocoral, Phenganax parrini, Sarcothelia sp., and Sympodium sp. After incubation of colonies for 1 h at elevated temperature, imaging and illumination (excitation 450–490 nm, emission 515–565 nm) were begun simultaneously. Formation of ROS largely corresponded to the onset of illumination. On the other hand, chlorophyll fluorescence (excitation 530–580 nm, emission 620–690 nm) did not conform to this pattern. This difference is consistent with the expected rates of reaction. Remarkably, treatment with the inhibitor 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) resulted in dramatically higher levels of light-induced ROS. Chlorophyll fluorescence was higher in the DCMU treatment but not significantly so. By controlling for variation between individual symbionts, however, DCMU produced significantly greater levels of chlorophyll fluorescence, indicating the expected greater reduction of photosystem II. A brief exposure to light and thermal stress produced a similar effect in all three species. In addition to ROS being initiated by light, these results indicate that: (1) a brief period of stress shifts photosystem redox state toward reduction, (2) photosystem II can donate electrons to oxygen when blocked with DCMU, and (3) chlorophyll fluorescence is highly variable among individuals of Symbiodinium. Imaging of individual symbionts in hospite thus provides a powerful method for understanding the initial steps of the cnidarian stress response.

The arthropod cuticle is a structurally diverse secretion that is largely composed of lipids, proteins, and α-chitin that function together in protection, prey capture, and as a skeletal framework for efficient and diverse means of locomotion. Aquatic, aerial, and terrestrial arthropods are well known to enrich their cuticles with trace elements that are proposed to strengthen or otherwise enhance specific regions of the exoskeleton. In this study, we provide evidence that whipscorpions (vinegaroons) enrich their exoskeleton with up to 14 trace elements. We use energy-dispersive x-ray spectroscopy (EDS) to provide the first comparisons of trace element composition in two species of Thelyphonida: Mastigoproctus giganteus (female) and Typopeltis dalyi (female and male). We analyzed the chemical composition of eight regions that have sensory, locomotory, taxonomic, protective, and/or predatory significance: the flagellum, prosoma, tarsal claws of the walking legs, tarsi of the antenniform legs, chelicerae, and the terminal three segments of the pedipalps. Our results reveal the presence of 14 trace elements across both species (12 elements in T. dalyi, 10 elements in M. giganteus), and only four elements are present at significant levels (≥1% weight): Si, Cl, Ca, and Zn. The flagellum, antenniform leg tarsi, and prosoma keel lack these elements in both species, while the chelicerae of all species are enriched with Ca, Zn, and Cl, and the tarsal claws are enriched with Zn. Significantly, we note the presence of Si in the prosomal carapace (but not the keel) of males and females of Typopeltis only, which appears to be the first evidence of this transition metal in the arthropod exoskeleton. We discuss the significance of these chemical enrichments in whipscorpions and provide hypotheses about their functional significance.
